摘 要:目的观察氨氯地平对肺腺癌A549细胞生长及上皮间质转化的影响,并初探其作用机制。方法将本实验中培养的肺腺癌A549细胞分空白对照组、二甲亚砜(DMSO)、氨氯地平组,采用CCK-8法检测细胞增殖抑制率;流式细胞仪检测细胞凋亡及周期变化;Western Blot法检测上皮间质转化相关蛋白表达。结果与空白对照组比较,氨氯地平对A549细胞具有显著抑制作用(P<0.05),IC50值为4.93μg/ml;氨氯地平对A549细胞具有促凋亡作用(P<0.05),且处理组G1期细胞明显增多(P<0.05),S期细胞明显减少(P<0.05);Western Blot及PCR方法检测发现Snail的蛋白和mRNA表达显著下调,E-cadherin的蛋白和mRNA表达显著上调(P<0.05)。结论氨氯地平可通过阻滞G1/S期细胞增殖来抑制A549细胞的增殖,促进细胞的凋亡,同时改变上皮间质转化靶基因Snail和E-cadherin的表达。Objective To observe the effects of amlodipine on the growth and epithelial mesenchymal transition of lung adenocarcinoma A549 cell lines,and to explore its mechanism. Methods The cultured lung adenocarcinoma A549 cells in this experiment were divided into a blank control group,a negative DMSO control group,a 0.625μg/ml amlodipine group,a 1.25μg/ml amlodipine group,a 2.50μg/ml amlodipine group,a 5.00μg/ml amlodipine group and a 10.00μg/ml amlodipine group.The inhibition rate of cell proliferation was detected by CCK-8 technique.The apoptosis and the cell cycle were detected by flow cytometry.The expression of epithelial mesenchymal transition related protein was detected by Western Blot technique.The measurements were recorded and the statistically comparative analysis was performed. Results By comparing with blank control group,Amlodipine had significant inhibitory effects on A549 cells in a dose dependent manner(P〈 0.05),the IC50 value was 4.93μg/ml.Amlodipine had the effects of promoting A549 cells apoptosis(P〈 0.05).The G1 phase cells of 1.25μg/ml and 2.50μg/ml groups were increased significantly(P〈 0.05),and S phase cells decreased significantly(P〈 0.05).The detection by Western Blot and PCR methods found that the expressions of protein and mRNA of Snail were significantly down regulated,and the protein and mRNA expressions of E-cadherin were significantly up-regulated(P〈 0.05). Conclusion Amlodipine can inhibit the proliferation of A549 cells,promote cell apoptosis and at the same time,change the expressions of epithelial mesenchymal transition target genes Snail and E-cadherin by blocking the proliferation of G1/S phase cells.
