摘 要:目的探讨山核桃叶提取物球松素对小鼠胚胎间充质干细胞C3H10T1/2成脂分化的影响。方法 MTS法检测不同浓度球松素处理的细胞成活率以界定适宜的浓度范围;油红O染色法结合异丙醇萃取法定性和定量分析球松素对细胞成脂分化的影响;GPO-PAP酶法检测球松素处理后细胞中甘油三酯含量的变化;荧光定量PCR和Western blot分别检测成脂分化中C/EBPα、PPARγ、FABP4 mRNA和相关蛋白的表达水平。结果 30μmol/L、50μmol/L球松素对细胞成脂分化有明显的抑制作用,且能显著减少甘油三酯堆积;10μmol/L、30μmol/L、50μmol/L球松素能明显抑制PPARγ、C/EBPαmRNA表达;5μmol/L、10μmol/L、30μmol/L、50μmol/L球松素能明显降低FABP4 mRNA表达;50μmol/L球松素能明显降低PPARγ、C/EBPα和FABP4蛋白表达。结论山核桃叶提取物球松素对C3H10T1/2细胞成脂分化具有抑制作用,可能与调控PPAR信号通路和相关基因表达有关。Objective To study the effects of pinostrobin extracted from Carya cathayensis Sarg Leaf on adipogenic differentiation of mouse embryonic mesenchymal stem cell C3H10T1/2. Methods The MTS was used to test the survival rate of cells treated with different concentrations of pinostrobin,so as to master the optimal concentration range. The adipogenic differentiation of cells was characterized by Oil Red O staining and isopropyl alcohol extraction qualitatively and quantitatively; the triglyceride content was detected by GPO-PAP enzymatic; the differentiation into adipocytes was also examined in the mRNA and the relational protein levels of PPARγ,C/EBPα and FABP4 by Q-PCR and Western blot respectively. Results Pinostrobin(30 μmol/L and 50 μmol/L) could significantly inhibit the adipogenic differentiation of Cell C3H10T1/2,and could also reduce the accumulation of triglycerid; pinostrobin could obviously decrease the expressions of PPARγ and C/EBPα mRNA(10 μmol/L,30 μmol/L and 50 μmol/L) and FABP4 mRNA(5 μmol/L,10 μmol/L,30 μmol/L and 50 μmol/L); pinostrobin(50 μmol/L) could decrease the expressions of PPARγ,C/EBPα and FABP4 protein. Conclusion It is suggested that pinostrobin from Carya cathayensis Sarg Leaf has a suppressive effect on adipogenic differentiation of Cell C3H10T1/2,and the mechanism may be associated with the regulation of PPAR signaling pathway and the gene expressions.
