哮喘大鼠肺组织Lyn激酶mRNA和蛋白表达的检测及意义

摘　　要：目的探讨Lyn激酶在哮喘发病机制中的作用。方法通过卵白蛋白致敏的哮喘大鼠模型,分别采用免疫组化法及荧光PCR法检测肺组织Lyn激酶mRNA和蛋白的表达。结果哮喘组Lyn激酶mRNA和蛋白的表达水平显著低于对照组(P<0.01);地塞米松组Lyn激酶mRNA的表达量与哮喘组差异无统计学意义(P>0.05),且显著低于对照组(P<0.01);地塞米松组Lyn激酶蛋白显著高于哮喘组且显著低于对照组(P<0.05)。肺组织Lyn激酶mRNA和蛋白的表达水平呈显著正相关(n=24,r=0.58,P<0.01)。结论哮喘大鼠肺组织Lyn激酶的表达降低,Lyn激酶在哮喘的发病机制中可能起了抑制气道炎症的作用,Lyn激酶抑制炎症的功能可以被地塞米松调节。Objective To investigate the potential roles of Lyn kinase in asthma pathogenesis. Methods The expression of Lyn kinase mRNA and protein in lung tissue was detected by immunohistochemistry and fluorescent PCR respectively in ovalbumin-sensitized asthma rat model. Results The expression levels of Lyn kinase mRNA and protein in the asthma group were significantly lower than those in the control group（P〈0. 01）. The expression of Lyn kinase mRNA in the dexamethasone group was copared with that in the asthma group,and the differences were not statistically significant（P〈0. 05）,and was significantly lower than that of the control group（P〈0. 01）; the kinetic acid protein of dexamethasone group was significantly higher than that of the asthma group but lower than that in the control group（P〈0. 05）. There was a significant positive correlation between the expression of Lyn kinase mRNA and protein in lung tissue（n = 24,r = 0. 58,P〈0. 01）. Conclusion The expression of Lyn kinase in lung tissue of asthmatic rats decreased. Lyn kinase may play a role in inhibiting airway inflammation in the pathogenesis of asthma. The anti-inflammatory function of Lyn kinase can be regulated by dexamethasone.
