摘  要:为了确定腹腔注射芥子碱降解菌1(sinapine-degrading bacteria 1,SDB1)对昆明小白鼠的安全性,试验分别采用革兰氏染色镜检法、活菌计数法、临床诊断、病理剖检和寇氏法对SDB1的特征、原菌液浓度、致病性以及SDB1对昆明小白鼠的半数致死量(LD_(50))进行了研究。结果表明:SDB1为中等大小、两端钝圆、散在或成对的革兰阴性杆菌。原菌液浓度为2.28×1010cfu/mL。腹腔注射SDB1后1~2小时,昆明小白鼠出现精神萎靡、食欲废绝、眼睛发炎、腹泻等临床表现;腹腔注射后10小时发生死亡;腹腔注射后12~24小时死亡达高峰,死亡数量与SDB1的腹腔注射量呈明显剂量-效应关系;耐过的昆明小白鼠2 d后不再发生死亡,且精神、食欲等逐步恢复正常。死亡昆明小白鼠主要病理剖检变化为肺脏肿大、瘀血、出血严重;胃肠充血、出血;肝脏、脾脏肿大。SDB1对昆明小白鼠的LD_(50)为3.80×10~8cfu,其95%可信限为2.96×10~8~4.88×10~8cfu。说明SDB1对昆明小白鼠有致病和致死性。The aim of the present study was to determine the satety ot sinapine-degrading bacteria 1 ( SDB1 ) rejected mtrapentoneally m Kun- ming mice. The classification, concentration of stock solution, the pathogenicity and LDs0of SDB1 in Kunming mice were studied by Gram stai- ning, viable counting, clinical diagnosis, pathological anatomy and Karber method.The results showed that the SDB1 wasa scattered or paired median-size gram negative bacillus with dull circle on both sides under the microscope. The viable count of the stock solution was 2.28x 1010 cfu/mL. About 1-2 hours after intraperitoneal injection of SDB1, the mice were poor spirit, anorexia, ophthalmitis, diarrhea. About 10 hours after injection, the mice began to die. About 12-24 hours after injection, the mortality reached the highest level.There was a signifi- cant dose-effect relationship between the death rate of the mice and the injection volume of SDB1. After 2 days, the mice did not die, and their spirits and appetite gradually returned to normal. The main pathological changes of the dead Kunming mice were pulmonary enlargement, blood stasis, severe bleeding, gastrointestinal congestion or hemorrhage, liver and spleen swelling.The LDso of SDB1 was 3.80× 108cfu and the confi- dence limit of 95% was 2.96x10s -4. 88×108cfu.lt was indicated that the SDB1 was pathogenic and fatal for Kunming mice.
