摘 要:目的:比较表柔比星(epirubicin hydrochloride)、奥沙利铂(oxaliplatin)、紫杉醇(taxol)、羟基喜树碱(10-hydroxycamptothecin)4种抗肿瘤药物对宫颈癌Si Ha细胞的体外抗肿瘤活性。方法:分别以1μmol/L、10μmol/L的带药培养基培养Si Ha细胞,通过MTT比色法测定4种药物对Si Ha细胞的体外抑制活性;通过形态学观察分析4种药物在1μmol/L、10μmol/L浓度下对细胞的作用。结果:4种药物对Si Ha细胞均表现出明显的抑制作用(P<0.05),其中表柔比星和紫杉醇在1μmol/L和10μmol/L、羟基喜树碱在10μmol/L时对Si Ha细胞的抑制率>70%,羟基喜树碱和奥沙利铂在1μmol/L时抑制率相对较低。形态学观察发现1μmol/L和10μmol/L表柔比星、紫杉醇以及10μmol/L羟基喜树碱处理时,细胞出现明显的皱缩破裂,表明药物的细胞毒性明显,1μmol/L和10μmol/L奥沙利铂及1μmol/L羟基喜树碱对细胞的影响主要表现在抑制细胞的增殖,细胞毒性相对较小。结论:表柔比星、紫杉醇以及高浓度的羟基喜树碱主要通过细胞毒性发挥抗肿瘤作用,而奥沙利铂和低浓度的羟基喜树碱通过抑制细胞增殖发挥抗肿瘤活性。Objective:To evaluate the antitumor activities of epirubicin hydrochloride, oxaliplatin, taxol and 10 -hydroxycamptotheein against cervical cancer Si- Ha cells in vitro. Methods:SiHa cells were cultivated in 1 μmol/L and 10 p-mol/L media and the Antieaneer activities in vitro were measured through MTT assay. Morphological observation was used for determining cells death. Results:The four agents showed obvious antieaneer activities against cervical cancer SiHa cells in vitro ( P 〈0.05). The inhibition rates of Epirubiein Hydmehloride and Taxol at 1 μmol/L and 10 μmol/L, and 10 - Hydmxyeamptotheein at 10 μmol/L were more than 70 %. Oxaliplatin and 10 - Hydroxyeamptotheein at 1 μmol/L were relatively low. Morphological observation showed that the treatments of Epirubiein Hydroehloride and Taxol at 1 μmol/L and 10 μmol/L, and 10 -Hydmxyeamptotheein at 10 μmol/L resulted in cell damage, showing the high eytotoxieity; Oxaliplatin at 1 μmol/ L and 10 μmol/L and 10 -Hydroxyeamptotheein at 1 μmol/L caused the proliferation of suppressor cells with low eytotoxieity. Conclusions:Epirubiein Hydroehlo- ride, Taxoi and 10 - Hydmxyeamptotheein at high concentration exert antieaneer activities mainly through eytotoxieity, while Oxaliplatin and 10 - Hydmxyeamptotheein at low concentration mainly through physiological pathway.
