摘 要:目的观察调衡方多糖对Lewis肺癌荷瘤小鼠红细胞补体受体1(CR1)功能及活性的影响,探讨该方多糖对荷瘤体红细胞作用的免疫调节机制。方法按照常规法在小鼠右腋皮下接种Lewis肺癌细胞,建立转移性肺癌模型;灌胃给药8d后,次日检测调衡方多糖对瘤体及脾脏、胸腺指数的影响;用红细胞免疫花环实验观察血液中红细胞免疫黏附肿瘤细胞的能力;红细胞免疫复合物花环实验观察荷瘤小鼠红细胞C3b受体的活性;比色法测定荷瘤小鼠红细胞膜唾液酸含量。结果调衡方多糖既可提高脾脏、胸腺指数及抑瘤作用,又可提升荷瘤小鼠红细胞免疫花环率;使荷瘤小鼠红细胞的活性与CR1受体的数量、荷瘤小鼠红细胞膜唾液酸含量增加。结论提示调衡方多糖发挥抑瘤与免疫调节作用的机制可能与其提升Lewis肺癌荷瘤小鼠红细胞膜上唾液酸的量有关,进而使CR1的数量与活性增加,使红细胞的黏附功能增强。Objective To investigate the immune regulation mechanism of Tiaoheng Prescription Polysaccharides (THPPS) towards tumor-bearing mice erythrocyte and the effects of THPPS on the function and activity of Lewis lung cancer tumor-bearing mice erythrocyte complement receptor 1(CR1). Methods The following experiments were implemented:establishing the metastatic lung cancer model by inoculating with Lewis lung carcinoma cells into the right armpit of mice subcutaneously according to the routine method;checking the weight of the tumor and the index of the spleen (thymus) by giving the THPPS after 8 days of oral administration;testing the RBC immune ability in the blood adhering to tumor cells by using red blood cell immune wreath experiment;the sialic acid level of red cell membrane in tumor-bearing mice was measured by colorimetric method. Results THPPS could increase the labeling index level in spleen or thymus,suppress the tumor production and increase the erythrocyte immune rate in tumor-bearing mice.It also showed that the THPPS could activate the red blood cell in tumor-bearing mice as well as raise the amount of CR1receptor and sialic acid level of erythrocyte membrane. Conclusion The function of THPPS in regulating tumor suppression and immune modulation may relate to the increasing activity level of CR1and immune adherence ability of red blood cell by THPPS in tumor-bearing mice.
