注射用艾司奥美拉唑钠对大鼠胃酸分泌作用的影响

摘　　要：目的:为获得国产注射用艾司奥美拉唑钠(艾速平)对大鼠胃酸分泌的药效学数据,比较其与原研药(耐信)在动物体内抑酸效果的差异。方法:本研究选取SD大鼠93只,随机分成6组:艾速平低剂量组(10 mg·kg-1·d-1)、艾速平高剂量组(50 mg·kg-1·d-1)、耐信低剂量组(10 mg·kg-1·d-1)、耐信高剂量组(50 mg·kg-1·d-1)、生理盐水组和空白组。于d 5和d 14,检测大鼠胃内p H值、胃蛋白酶及胃泌素蛋白表达水平。结果:国产注射用艾司奥美拉唑钠对比原研用药d 5及d 14后,均可有效抑制大鼠胃酸分泌,两组大鼠胃内p H值水平、胃蛋白酶及胃泌素表达水平均一致,无统计学差异。结论:国产注射用艾司奥美拉唑钠可有效抑制大鼠胃酸分泌,具有强而持久的抑酸效果。Objective： To compare the effects of generic（ Aisuping） and original intravenous esomeprazole（ Nexium） on gasctric acid secretion. Methods： Ninety-three SD rats were randomly divided into six groups,including Aisupinglow-dose group（ 10 mg·kg^-1·d^-1）,Aisuping high-dosegroup（ 50 mg·kg^-1·d^-1）,Nexiumlow-dose group（ 10 mg·kg^-1·d^-1）,Nexiumhigh-dose group（ 50 mg·kg^-1·d^-1）,saline group and blank group.Then,the secretion levels of gastric acid,pepsin and gastrin were detected on day 5 and day 14 after the multiple dose administration. Results： There was no significant difference in the secretion levels of gastric acid,pepsin and gastrin between Aisupingand Nexiumgroups on day 5 and day 14. Both Aisupingand Nexiumcould effectively inhibit gastric acid secretion in rats. Conclusion： Generic esomeprazole for injection has long and persistent gastric acid inhibitory effect in rats,which is equivalent to original esomeprazole.
