摘 要:目的优选白背叶总黄酮类成分的提取、纯化工艺条件,为白背叶中总黄酮工业生产提供科学依据。方法以白背叶总黄酮含量为指标,采用单因素试验及正交实验设计方法 ,确立白背叶总黄酮类成分最佳提取工艺;以总黄酮纯度和收率为指标,采用聚酰胺树脂(80~100目)考察样品吸附浓度、吸附量、吸附液p H、洗脱溶剂等因素,确定其最佳纯化工艺。结果白背叶最佳提取工艺为50%乙醇提取,料液比为1∶70,加热回流提取2次,每次1 h,提取得到白背叶总黄酮含量为4.040%;调节白背叶提取液p H4,浓度为4 mg/ml,上样量为1 BV,先用8/5 BV水洗脱除杂,再用90%乙醇洗脱至澄清。纯化后白背叶总黄酮含量为50.32%,收率为92.74%。结论该工艺简便快捷、成本低、毒性小、选材可重复使用,适合工业化生产。Objective To study the extraction and purification process of Mallotus apelta. And provided reasonable method for total flavonoid industrial production in Mallotus apelta. Methods The content of total flavonoids is used as an indicator. Using single factortest and orthogonal experiment design method to establish the optimum extraction process of total flavonoids in Mallotus apelta; The content of total flavonoids is used as an indicator. Adsorption duration, capacity, p H and alcohol concentration are investigated by polyamide resin(80 ~ 100 mesh) with total flavones product purity and yield as index. And the best purification process is determined.Results The optimal extraction process conditions are 50 % ethanol extraction, the ratio of material to liquid is 1 : 70, extraction for 2 times and 2 h for each time are optimal.The content of total flavonoids in Mallotus apeltais 4.040 %. The extraction process conditions are p H value of 4, sample mass concentration of 4 mg/ml, The loading quantities is 1 BV. The sample is eluted by water with 8/5 BV and 90 % ethanol elution to clarify.The content of total flavonoids in Mallotus apelta is 50.32 % after purification and the yield is 92.74 %. ConclusionThe technology is convenient and rapidy, low cost, small toxicity, materials can be reused. Material can be reused for industrial production. It is suitable for industrial production.
