摘  要:目的采用金属络合的方法分离纯化苦碟子中的黄酮类活性成分。方法以Mg^(2+)、Ca^(2+)、Zn^(2+)作为络合剂,以木犀草素-7-O-β-D-葡萄糖醛酸苷(Luteolin-7-O-β-D-glucuronide,LGU)为指标成分,HPLC-UV法测定LGU的含量和转移率,考察不同pH(8.0、9.0、9.5、10.0、11.0)条件下各金属离子对苦碟子黄酮的纯化效果。结果 pH=9.5时,Mg^(2+)络合物中LGU含量最高,与纯化前的醇提取物相比,含量提高5倍达到12.1%。结论采用金属络合法纯化苦碟子黄酮是一种简单、有效的纯化方法。Objective To study the purification technology of flavonoid from the Ixeris sonchifolia(EIS) by metal complex method. Methods Luteolin-7-O-β-D-glucuronide(LGU) as the key parameters, the contents and transfer rates in different metal complex(Mg2+, Ca2+ or Zn2+) prepared at various p H of 8.0, 9.0, 9.5, 10.0 and 11.0 are investigated by HPLC-UV method. Results The results indicated that the complexation by Mg2+ at pH 9.5 is the optimum. With this optimum condition, the content of LGU in Mg2+ complex is 12.1 %, about 5 times as much that in EIS. Conclusion Purification of flavonoid Ixeris sonchifolia by metal chelating method is simple and effective.
