摘 要:目的研究用12种不同型号大孔树脂富集纯化重楼中重楼皂苷Ⅱ、纤细薯蓣皂苷、重楼皂苷I的最佳工艺。方法采用HPLC法测定重楼皂苷Ⅱ、纤细薯蓣皂苷、重楼皂苷Ⅰ的含量,比较在12种不同型号大孔树脂下3种单体的静态吸附量和解吸率,并根据动态吸附和解析实验,确定最佳的吸附洗脱工艺。结果 ADS-17树脂对3种重楼皂苷单体的吸附能力和解吸率较强,40%乙醇用于除杂,洗脱8个柱体积,90%乙醇用于富集皂苷单体,洗脱15个柱体积,浸膏得率为14.6%,浸膏中重楼皂苷Ⅱ、纤细薯蓣皂苷、重楼皂苷Ⅰ的含量分别为64.14、9.50、108.37 mg·g^(-1),转移率分别为87.67%、91.63%、93.98%。结论该工艺能有效富集纯化重楼中重楼皂苷Ⅱ、纤细薯蓣皂苷和重楼皂苷Ⅰ。Objective To study the optimal process for gathering and purifying three steroidal saponins from Paris Polyphylla with twelve kinds of macroporous resins.Methods Twelve kinds of macroporous resins were investigated through adsorption and desorption tests,and the contents of Paris saponinⅡ,gracillin and Paris saponinⅠwere analyzed by HPLC.Results ADS-17 resin showed the maximum effectiveness and was thus selected for subsequent tests,in which 40% ethanol(8 BV)was used to remove the undesired constituents and 90%ethanol(15 BV)to elute the targets.The yield of the enriched extract was 14.6%,the contents of Paris saponinⅡ,gracillin and Paris saponin I were 64.14,9.50 and 108.37 mg·g-1 respectively,and their diversion rates were87.67%,91.63% and 93.98% respectively.Conclusion This process can effectively purify three steroidal saponins fromParis Polyphylla.
