摘 要:TAC1基因在哺乳动物神经系统中广泛表达,其编码蛋白参与季节性生殖活动的调节。该研究对杜泊羊速激肽(Tachykinin,TK)基因序列进行了克隆及生物信息学分析。结果表明:克隆出杜泊羊TAC1基因组序列,得到3个克隆片段。通过生物信息学分析,克隆片段一的长度为860 bp,经BLAST对比,与云南黑山羊第4号染色体相似度达93%,基因片段缺失4%;克隆片段二的长度为881 bp,经BLAST对比,与云南黑山羊第4号染色体相似度达100%,没有基因片段的缺失;克隆片段三的长度为846 bp,经BLAST对比,与云南黑山羊第4号染色体相似度达98%,没有基因片段的缺失,但有3个突变位点,分别为位于85 bp处的G/C突变、位于88 bp处和629 bp处的G/A突变、位于108 bp处和290 bp处的T/C突变。杜泊羊TAC1基因开放阅读框全长423 bp,共编码140个氨基酸。与Gen Bank已发表的其他动物TAC1基因参考序列进行比较,结果显示杜泊羊与绵羊、瘤牛、野猪、猕猴、人和大熊猫的TAC1基因同源性分别为98.9%、97.3%、88.1%、85.8%、85.0%、84.5%。通过蛋白质结构预测,发现杜泊羊TAC1基因编码蛋白没有跨膜结构域。TAC1 gene is widely expressed in many nervous tissues in mammalians,which participates in regulating the seasonal reproduction in animals. Tachykinin gene were cloned and bioinformatics analysis were performed in Dorper sheep. The results showed that TAC1 gene of Dorper sheep was cloned successfully and three cloned fragments were got.The length of the first cloned fragment was 860 bp,which had 93% similarity with Yunnan black goat chromosome No. 4 and 4% deletion by BLAST analysis;The length of second cloned fragment was 881 bp,which had 100% similarity to Yunnan black goat chromosome No. 4 by BLAST analysis;The length of the third cloned fragment was 846 bp,which had 98% similarity to Yunnan black goat chromosome No 4.There were three potential mutations in the third fragment,including a G/C mutation at 85 bp,two G/A mutations at 88 bp and 629 bp,and tow T/C mutation at 108 bp and 290 bp,respectively. The open reading frame of the TAC1 gene in Dorper sheep was 423 bp and encoded a total of 140 amino acids. Compared Dorper sheep with the published TAC1 gene reference sequence of other animals published in Gen Bank,the results showed that the TAC1 gene of Ovis aries,Bos indicus,Sus scrofa,Macaca nemestrina,Homo sapiens,Ailuropoda melanoleuca the sources were 98.9%,97.3%,88.1%,85.8%,85.0%,84.5%.Through the prediction of protein structure,it was found that TAC1 of the Dorper sheep had no transmembrane domain.
