疏肝补肾法对慢性应激肝郁证模型小鼠子宫内膜容受性的影响

摘　　要：目的:观察疏肝补肾法对慢性应激肝郁证模型小鼠子宫内膜容受性的影响。方法:105只6周龄KM雌性小鼠随机分为空白组、模型组、逍遥丸组、左归丸组、逍遥丸加左归丸组、疏肝补肾中药高剂量组、疏肝补肾中药低剂量组。除空白组外,其余各组均接受21 d的应激刺激,各组给药按10 m L·kg-1,每日1次。行为测试并合笼后,以见到阴道栓作为受孕第1天,孕后小鼠持续接受慢性应激刺激,于孕第5天处死,检测各组小鼠血清雌二醇(estradiol,E2)、孕酮(progesterone,P)、绒毛膜促性腺激素(human chorionic gonadotropin,HCG)及子宫内膜组织中血管内皮生长因子(vascular endothelial growth factor,VEGF)的水平。结果:(1)与模型组比较,空白组、逍遥丸加左归丸组、疏肝补肾中药高剂量组小鼠体质量增长较快(P<0.05);(2)空白组、疏肝补肾中药高剂量组中央格停留时间显著短于模型组(P<0.05);(3)空白组、逍遥丸加左归丸组、疏肝补肾中药高剂量组血清E2、P、HCG及子宫内膜组织中VEGF水平显著高于模型组(P<0.05);(4)空白组、逍遥丸加左归丸组及疏肝补肾中药高剂量组的阴道见栓率、胚胎数明显高于模型组(P<0.05)。结论:(1)慢性应激可导致小鼠子宫内膜容受性降低;(2)疏肝补肾方能改善慢性应激雌性小鼠的应激状态,调节慢性应激雌性小鼠的性激素水平,提高血清中E2、P及内膜VEGF表达,促进胎盘分泌HCG,进而改善子宫内膜微环境。Objective： To observe the effect of Shugan Bushen Method on endometrial receptivity in mice with chronic stress-induced Liver stagnation syndrome. Methods： A total of 105 6-week-old KM female mice were randomly divided into blank group,model group,Xiaoyao Pill group,Zuogui Pill group,Xiaoyao Pill plus Zuogui Pill group,Shugan Bushen Chinese medicine high-dose group and Shugan Bushen Chinese medicine low-dosa group. Except for the blank group,the other groups all received 21 d stress stimulation,and each group was administered with 10 m L·kg^（-1） once a day. After the behavior test and being putting in the same cage,the vaginal suppository was used as the index of the first day of conception. The mice continued to receive chronic stress stimulation after pregnancy,and were sacrificed on the 5 th day of pregnancy. Serum estradiol（ estradiol,E2） was detected in each group. Progesterone（ P）,human chorionic gonadotropin（ HCG）,and levels of vascular endothelial growth factor（ VEGF） in endometrial tissue of all groups were detected. Results：① Compared with that of the model group,the body weight of the blank group,Xiaoyao Pill plus Zuogui Pill group and Shugan Bushen high-dose group increased significantly（ P〈0. 05）;② The retention time of the central compartment of rats in blank group and Shugan Bushen Chinese medicine high-dose group was significantly shorter than that of the model group（ P〈0. 05）.③ The levels of VEGF in the serum E2,P,HCG and endometrial tissues of the blank group,Xiaoyao Pill plus Zuogui Pill group and Shugan Bushen high-dose group were significantly higher than that of the model group（ P〈0. 05）;④ the vaginal thrombosis rate and the number of embryos of the blank group,Xiaoyao Pill plus Zuogui Pill group and Shugan Bushen high-dose group were significantly higher than those in the model group（ P〈0. 05）. Conclusion：①Chronic stress can lead to the reduction of endometrial receptivity in mice;② Shugan Bushen method can improve the stress
