摘 要:目的:观察雷公藤甲素、雷公藤内酯酮、雷酚内酯、雷公藤次碱、雷公藤红素及雷公藤内酯甲对仓鼠卵巢细胞凋亡的浓度及时间效应。方法:采用体外细胞培养,用不同质量浓度的雷公藤单体干预及相同质量浓度干预不同时间,显微镜下观察细胞形态及数量变化,Annexin V-FITC/PI法检测细胞凋亡率及蛋白印迹法检测细胞ERK磷酸化及c-fos表达水平。结果:不同单体干预的细胞凋亡率、ERK磷酸化及c-fos表达水平不同,以雷公藤甲素、雷酚内酯、雷公藤红素、雷公藤内酯甲最显著。雷公藤甲素质量浓度在30μg·L^(-1)时细胞凋亡率最高,其后质量浓度增加,细胞凋亡率并不增加;雷酚内酯干预后细胞凋亡呈浓度效应即随质量浓度增加细胞凋亡率升高;雷酚内酯、雷公藤红素、雷公藤内酯甲干预后随干预时间延长细胞ERK磷酸化及c-fos表达增加。结论:不同雷公藤单体引起细胞凋亡的质量浓度有差异,雷公藤甲素促进细胞凋亡的最佳质量浓度是30μg·L^(-1),而雷酚内酯在10~200μg·L^(-1)范围内随质量浓度增加,引起细胞凋亡增加的现象。此外雷酚内酯、雷公藤红素、雷公藤内酯甲引起细胞凋亡相关蛋白的表达有时间效应。Objective: To observe the concentration and time effect of triptolide,tripterygium lactone,ray phenol lactone,wilforine,celastrol,and wilforlide A HPLC on ovarian cell apoptosis in hamsters. Methods: In vitro cell culture was used to observe the changes of cell morphology and quantity under different microscopes with different concentrations of tripterygium wilfordii and then with the same concentration of tripterygium wilfordii for different periods of time. The apoptosis rate was detected by Annexin V-FITC/PI method and the cell ERK Phosphorylation and c-fos expression levels were detected by Western blotting. Results: Apoptosis rate,ERK phosphorylation and c-fos expression levels were different in different monomer interventions,in which the effects of triptolide,ray phenol lactone,celastrol,and wilforlide A HPLC were the most prominent. Triptolide had the highest apoptotic rate at 30μg·L-1 concentration,and then when the concentration increased the apoptosis rate did not increase. After the treatment with Ray phenol lactone,the concentration of apoptosis increased,and the apoptosis always increased with concentration. Ray phenol lactone,celastrol,and wilforlide A HPLC increased the phosphorylation of ERK and the expression of c-fos in the cells with the intervention time. Conclusion: Different tripterygium wilfordii that induce the cell apoptosis is different in concentration. The optimal concentration of triptolide to promote apoptosis is 30 μg·L-1,and the content of resphenol lactone is in the range of 10-200μg·L-1. As the concentration increases,it causes an increase in apoptosis. In addition,ray phenol lactone,celastrol,and wilforlide A HPLC have a time effect on the expression of apoptosis-related proteins.
