红景天苷对高同型半胱氨酸诱导的小鼠膀胱胶原沉积及SGK1蛋白表达的影响

摘　　要：目的:观察红景天苷对高同型半胱氨酸诱导的小鼠膀胱沉积胶原的影响。方法:40只清洁级BABLc雄性小鼠适应饲养7 d后,随机分为空白对照组、模型组(1%高蛋氨酸饮水8周)、红景天苷25 mg·kg^(-1)组(0.01%的含红景天苷饮食喂养16周,后给予1%高蛋氨酸饮水8周)和红景天苷50 mg·kg^(-1)组(0.02%的含红景天苷饮食喂养16周,后给予1%高蛋氨酸饮水8周),每组10只。取膀胱组织,Masson染色检测胶原沉积;Western Blot法检测纤维化相关蛋白-血清和糖皮质激素诱导蛋白激酶1(Serum and glucocorticoid-induced protein kinase,SGK1)表达和免疫组化染色检测膀胱组织白介素-1β(Interleukin-1β,IL-1β)蛋白表达。结果:与模型组相比,红景天苷50 mg·kg^(-1)可显著抑制高蛋氨酸诱导的膀胱组织胶原沉积和纤维化相关蛋白SGK1及膀胱组织炎症因子IL-1β的表达(P<0.05)。结论:红景天苷可减轻高同型半胱氨酸诱导的小鼠膀胱胶原沉积和SGK1蛋白表达,同时抑制膀胱组织IL-1β蛋白表达。Objective： To study the effect of salidroside on cystic collagen deposition induced by hyperhomocysteinemia in mice. Methods： After one-week adaptive feeding, 40 male BABLc mice were randomly divided into the blank control group, the model group, （ 1% high methionine drinking water for 8 weeks） , salidroside at low （25 mg · kg-1 , 0.01% salidroside diet for 16 weeks, then given 1% high metbionine drinking water for 8 weeks） and high dose groups （50 mg· kg-1, 0.02% salidroside diet for 16 weeks, then given 1% high methionine drinking water for 8 weeks） with 10 ones in each. After the experiment, the mice were sacri- ficed by decapitation to collect the bladder tissues. Masson staining was used to detect the collagen deposition, Western blot was ap- plied to assess serum and glucocorticoid-induced protein kinase （SGK1） protein level, and immunohistochemistry was used to examine the expression of interleukin-1β （IL-1β）. Results： Compared with the model group, salidroside high dose could inhibit the methionine induced collagen deposition and the expression of SGK1 and IL-1β（P 〈 0.05）. Conclusion： Salidroside may attenuate cystic collagen deposition induced by hyperhomocysteinemia and reduce the expression of SGK1 and IL-1β.
