摘　　要：目的研究新疆家蚕抗菌肽Cecropin XJ抗肝癌细胞BEL-7402的功能并探讨其机制。方法分别利用CCK-8和体内移植瘤模型检测Cecropin XJ抑制肝癌细胞生长的功能,检测其对正常肝脏细胞(LO2细胞)和小鼠血液系统的影响,并通过Western blotting检测凋亡相关蛋白表达情况。结果 Cecropin XJ可显著抑制BEL-7402细胞的增殖和体内移植瘤的生长,对正常肝脏细胞和小鼠血液系统无明显影响,抑瘤率为36.7%,Western blotting结果显示Cecropin XJ可以显著上调Bax/Bcl-2的表达,促进Cleaved-Casp3的表达。结论Cecropin XJ具有抑制肝癌BEL-7402细胞增殖作用,并且无明显毒副作用,其机制与诱导肿瘤细胞凋亡相关。Objective： To study the effect and mechanism of Cecropin XJ on anti-hepatoma carcinoma cell BEL-7402. Methods： CCK-8 assay and xenografts were applied to detect Cecropin XJ inhibiting the proliferation of liver cancer. Meanwhile,its effects on normal liver cells and mice blood system were detected. The expression of apoptosis-associated proteins was investigated with Western blot analysis. Results： Cecropin XJ remarkably inhibited the proliferation of BEL-7402 cells and the growth of transplanted tumor,with no significant effect on normal liver cells and mice blood system. The tumor-inhibition rate was 36.7%. Western Blot analysis showed that Cecropin XJ significantly increased the ratio of Bax/Bcl-2 and cleaved-Casp3. Conclusion： Cecropin XJ can inhibit the proliferation of BEL-7402 cells without obvious side effects,and its mechanism is related to inducing apoptosis of tumor cells.
