摘 要:目的探究四环素抗纳米细菌(nanobacteria,NB)治疗对人肾小管上皮细胞HK-2损伤及晶体滞留的影响,为泌尿系结石的预防和治疗提供新的思路。方法将HK-2细胞分为3组:NB组、四环素干扰组、空白对照组。将人肾小管上皮细胞HK-2细胞分别与各组作用物共培养6 h,12 h和24 h后,用透射电子显微镜观察HK-2细胞的形态学变化;加入COM晶体后用激光共聚焦显微镜观察各组细胞的晶体黏附情况。结果电镜可见:NB组HK-2细胞膜局部区域结构破损,微绒毛结构稍紊乱,局部绒毛消失,细胞核畸形,细胞核外形不规则,呈双核或两分叶,部分线粒体结构轻度肿胀,四环素组HK-2细胞膜结构完整,部分线粒体结构肿胀不明显。激光共聚焦显微镜观察结果显示:随着共培养时间延长,NB组细胞的晶体黏附量持续增加。各时间点,四环素干扰组中晶体黏附量均显著少于NB组。结论 NB可能通过损伤HK-2细胞导致晶体滞留而引发肾结石的形成,并且损伤水平和晶体黏附量与NB作用时间成正比。而四环素可能通过抗NB作用减少HK-2细胞的损伤及晶体滞留。Objective To explore the effect of tetracycline-resistant nanobacteria (NB) treatment on HK-2 injury and crystal retention in human renal tubular epithelial cells,so as to provide new ideas for the prevention and treatment of urolithiasis. Methods HK-2 cells were divided into 3 groups,NB group,tetracycline interference group and blank control group. The HK-2 cells of human renal tubular epithelial cells were co-cultured with the substrates for 6 h,12 h and 24 h respectively. The morphological changes of HK-2 cells were observed by transmission electron microscope. After adding COM crystals,the adhesion of cells in each group was observed by laser scanning confocal microscope. Results Electron microscopy revealed that the HK-2 cell membrane in NB group was partially damaged,the microvilli structure was slightly disordered,the local villi disappeared,the nucleus was malformed,and the cell nucleus was irregular in appearance. The nucleus was double or two-leaf. Some mitochondria were slightly swollen. Confocal laser scanning microscopy results showed that with the prolonged co- culture time,the cell adhesion of NB group continued to increase. The crystal adhesion in the tetracycline interference group was significantly lower than that in the NB group at each time point. Conclusion NB may lead to the formation of kidney stones by damaging the HK-2 cells and causing the retention of the crystals,and the damage degree and the amount of crystal adhesion are proportional to the time of action of NB. Tetracycline may reduce HK-2 cell damage and crystal retention by anti-NB effect.
