丹溪痛风胶囊对胶原诱导性关节炎大鼠RORγt和Foxp3蛋白表达的影响

摘　　要：目的观察丹溪痛风胶囊对胶原诱导性关节炎(CIA)大鼠的脾组织淋巴细胞中RORγt和Foxp3蛋白表达的影响。方法研究地点为黑龙江中医药大学实验中心,2015年2月—10月选取SD大鼠30只,采用牛Ⅱ型胶原诱导法制备CIA大鼠模型,随机分成模型组、丹溪痛风胶囊组;另设正常对照组,每组10只。分别连续灌胃给予相应药物4 W。检测各组大鼠足趾肿胀度和关节炎指数,Western blot分析脾组织淋巴细胞中RORγt和Foxp3蛋白表达。结果模型组RORγt蛋白表达明显升高,Foxp3蛋白表达明显降低,分别为1.724±0.067和0.418±0.043,与对照组相比较(P<0.01);丹溪痛风胶囊组大鼠RORγt蛋白表达明显降低,Foxp3蛋白表达明显升高,分别为0.621±0.073和0.976±0.067,与模型组相比较(P<0.01)。结论丹溪痛风胶囊可以通过调节Th17与Treg细胞的特异转录因子RORγt和Foxp3的平衡,从而对类风湿性关节炎起到治疗的作用。Objective To observe the expression of the spleen RORγt and Foxp3 protein in rat collagen- induced arthritis （ClA） subjected TCM Danxi＇s gout capsule intervention. Methods In author＇s Experiment Center,Heilongjiang University of TCM from February to October 2015yr. The ClA model in 30 SD rats were established and randomly divided into normal group,model control group,Danxi＇s gout capsule treatment group,10 in each. Danxi＇s gout capsule treatment group was treated with Danxi＇s gout capsule by garage,while the normal group and model control group were fed with the same amount of saline. Four weeks after the treatment,observed the changes of the toe swelling degree and arthritis index.The lymphocytes were extracted from spleen,the expression of RORγt and Foxp3 protein were detected by Western blot. Results Compared with the normal group,the ROR3,t protein levels significantly increased（1.724±0.067）,Foxp3 protein levels decreased（0.418±0.043）in the model control group （P〈0.01）. Compared with the model control group,the ROR3,t protein levels significantly decreased（0.621±0.073）,Foxp3 protein levels increased（0.976±0.067） in the Danxi＇s gout capsule treatment group （P〈0.01）. Conclusion Danxi＇s gout capsule can reduce the expression of the spleen RORγt and Foxp3 protein, balance the Th17 and Treg cell specific transcription factor RORγt and Foxp3,and improve the immunity function of ClA rats.
