摘 要:目的:探讨分析玉米须总皂苷对高脂饮食喂养的肥胖大鼠棕色脂肪组织中解偶联蛋白-1(UCP1)和PR结构域蛋白16(PRDM16)表达的影响。方法:雄性健康清洁级SD大鼠共40只,在适应性喂养1周后随机分为正常对照组、模型对照组以及玉米须总皂苷低剂量和高剂量干预组,每组10只,给予高脂饮食12周后构建肥胖大鼠模型,治疗时玉米须总皂苷低和高剂量组分别给予玉米须总皂苷100、200 mg·kg^-1·d^-1灌胃,正常对照组和模型对照组给予1 m L·kg^-1·d^-1生理盐水灌胃,治疗12周后称重、处死大鼠,收集血清和棕色脂肪组织,检测血清血脂指标,HE染色检测棕色脂肪组织病理学,Real-Time PCR和Western Blot检测棕色脂肪组织中UCP1和PRDM16的m RNA和蛋白表达水平。结果:与正常对照组相比,模型对照组大鼠体重明显升高(P〈0.05),棕色脂肪组织重量和棕色脂肪组织重量/大鼠体质量明显降低(P〈0.05),与模型组相比,玉米须总皂苷治疗后,大鼠体质量明显降低(P〈0.05),棕色脂肪组织重量和棕色脂肪组织重量/大鼠体质量明显升高(P〈0.05);与正常对照组相比,模型对照组血清葡萄糖、甘油三酯、总胆固醇和HDL-C明显升高(P〈0.05),LDL-C明显降低(P〈0.05);与模型对照组相比,玉米须总皂苷治疗后,血清葡萄糖、甘油三酯水平、总胆固醇和HDL-C降低(P〈0.05),LDL-C明显升高(P〈0.05);与正常对照组相比,模型对照组棕色脂肪细胞分布明显减少,脂肪空泡数量明显增加,而玉米须总皂苷治疗后,脂肪空泡数量明显降低;与正常对照组相比,模型对照组大鼠棕色脂肪组织中UCP1和PRDM16的m RNA和蛋白表达明显降低(P〈0.05),玉米须总皂苷治疗后表达明显升高(P〈0.05)。结论:玉米须总皂苷治疗12周可以明显降低肥胖大鼠体质量和棕色脂肪组织中的相对含量,这可能与其可以降低棕色脂肪组织中UCP1和PRObjective:To investigate the effection of total saponins of maize on the expression of uncoupling protein-1(UCP-1)and PR domain protein 16(PRDM16)in brown adipose tissue of obese rats. Methods:Forty healthy and male SD rats were randomly divided into the normal group,model group and the total saponins of maize low and high dose intervention group,The rats in the normal and the model group were given 1 m L·kg^-1·d^-1 saline,low and high dose group were given total saponins of maize 100 mg·kg^-1·d^-1 and 200 mg·kg^-1·d^-1,after 12 weeks of treatment,rats were sacrificed and serum and brown adipose tissue were collected. Serum lipids were measured. Meanwhile,HE staining was used to detect brown adipose tissue pathology. Real-Time PCR and Western Blot were used to detect the expression of UCP1 and PRDM16 m RNA and protein in adipose tissue. Results:Compared with the normal group,the body weight in the model group was significantly increased(P〈0.05),the weight of brown adipose tissue and the weight of brown adipose tissue/body weight were significantly decreased(P〈0.05),compared with the model group(P〈0.05),the weight of the brown adipose tissue and the weight of the brown adipose tissue were significantly increased(P〈0.05);Compared with the normal group,LDL-C was significantly decreased(P〈0.05),triglyceride level,total cholesterol and HDL-C were significantly increased(P〈0.05),Compared with the model control group,after the total saponins of maize treated,serum and triglyceride level,total cholesterol and HDL-C were significantly decreased(P〈0.05),LDL-C was significantly increased(P〈0.05). Compared with the normal group,the distribution of brown fat cells in the model control group was significantly increased,and after treatment were decreased(P〈0.05). Compared with the normal group,the m RNA and protein expression of UCP1 and PRDM16 in the brown adipose tissue of the model control group were significantly lower than those in the control group�
