摘 要:目的:优选肝乐颗粒的水提工艺。方法:以绿原酸含量、芍药苷含量、柚皮苷含量、新橙皮苷含量及干浸膏得率为评价指标,利用L9(34)正交试验法,考察加水量、提取时间、提取次数及浸泡时间对肝乐颗粒水提工艺的影响。结果:肝乐颗粒最佳水提工艺为:药材加8倍量水,浸泡时间为2 h,水煮提取3次,每次1 h。按照优选出的最佳提取工艺进行验证试验,得到4种指标成分的含量分别为绿原酸(0.5236±0.0054)mg·g^-1,RSD为1.50%;芍药苷(4.257±0.0035)mg·g^-1,RSD为1.73%;柚皮苷(6.638±0.0026)mg·g^-1,RSD为1.87%;新橙皮苷(3.628±0.0062)mg·g^-1,RSD为0.99%;干浸膏得率为23.13%±0.36%,RSD为1.71%。结论:优选出的水提工艺科学合理、稳定可行,为肝乐颗粒的大规模生产提供了更加可靠的实验依据。Objective:To optimize water-extraction technology of Ganle Granule. Methods:The optimized extracting condition of Ganle Granule were investigated by determining the dried extract,the content of chlorogenic acid,the content of paeoniflorin,naringin and neohesperidin. Four factors,water volume,decocting time,extracting times,and soaking time were considered to effect the extracting efficiency of active component. Results:The reasonable extracting conditions were 8 times water for medicinal materials,2 h for soaking,decocting for three times with water for 1 hour each time. According to the optimal extraction process,the content of the four indexes were obtained as follows:chlorogenic acid(0.5236±0.0054)mg·g^-1,RSD 1.50%;paeoniflorin(4.257±0.0035)mg·g^-1,RSD 1.37%;naringin(6.638±0.0026)mg·g^-1,RSD 1.87%;neohesperidin(3.628±0.0062)mg·g^-1,RSD 0.99%;dry extract yield was 23.13%±0.36%,RSD 1.71%. Conclusion:It shows that the optimal water extraction process is scientifically reasonable,stable and feasible,which provides a reliable experimental basis for large-scale production of Ganle Granule.
