HIF-1α慢病毒和小孔径髓芯减压治疗兔早期激素性股骨头坏死的研究

摘　　要：目的构建低氧诱导因子(HIF-1α)慢病毒载体,并讨论其治疗早期激素性股骨头坏死的疗效。方法构建兔早期激素性股骨头坏死模型,按照不同处理进行分组:A组5只(HIF-1α慢病毒载体将目的基因载入兔早期激素性股骨头坏死模型组织细胞基因组中并进行小孔径髓芯减压处理),B组5只(仅进行小孔径髓芯减压处理)、C组2只(不做任何处理),并通过MRI、X线、CT影像检测评价3组兔激素性股骨头坏死后的修复情况以及在不同处理后不同时间点局部HIF-1α和其受体的含量。结果 HIF-1α慢病毒病毒滴度为4×106 TU/mL;8w后A组兔激素性股骨头坏死后的修复显著优于B组和C组。结论成功构建HIF-1α慢病毒载体;HIF-1α慢病毒对早期激素性股骨头坏死有一定疗效;HIF-1α参与早期激素性股骨头坏死组织的修复机制。Objective Construction of HIF-1α lentiviral vector,and to discuss its efficacy in the treatment of early steroid-induced femoral head necrosis.Methods Rabbit model of early steroid-induced femoral head necrosis was constructed,and grouped according to different processing.Group A：The HIF-1α lentiviral vector was used to carry the target gene into the genome of the rabbit model of early steroid-induced femoral head necrosis and perform small pore core decompression.Group B：Only small pore core core decompression treatment.Group C：the normal control group without any treatment.There were 5 rabbits in group A and B,respectivily,and 2 rabbits in group C.MRI,X-ray,CT images were used to evaluate the repair of steroid-induced femoral head necrosis in 3 rabbits.Results The HIF-1α lentivirus virus titer was4×106 TU/mL.At 8 weeks,the repair of steroid femoral head necrosis in group A was significantly better than that in group B and group C.Conclusion Successful construction of HIF-1α lentiviral vector.HIF-1α lentivirus has a certain effect on early steroid-induced femoral head necrosis.HIF-1α is involved in the repair mechanism of early steroid-induced necrosis of femoral head.
