摘  要:目的研究西洛他唑对大鼠心室肌细胞瞬间外向钾电流(Ito)、L型钙离子流(ICa-L)的影响,探讨其可能有的抗心律失常作用。方法将雄性SD大鼠随机分为对照组和观察组(西洛他唑组),喂养3~4周。将大鼠右心室肌单细胞使用胶原酶Ⅱ分离出单个细胞,采用全细胞膜片钳技术记录右心室肌细胞Ito及ICa-L离子通道电流。结果 50μmol·L^(-1)西洛他唑加入对照组细胞外灌流液,Ito幅值由加药前的(21. 55±2. 61) p A/p F降至(5. 36±2. 16) p A/p F,P <0. 05; 2μmol·L^(-1)西洛他唑加入对照组细胞外灌流液,Ito幅值由加药前的(18. 64±7. 89) p A/p F降至(7. 63±1. 78) p A/p F,P <0. 05;药物洗脱后Ito均有不同程度的恢复。而对比两组大鼠的Ito及L型钙离子流(ICa-L)的电流密度无明显变化,差异无统计学意义(P> 0. 05)。结论急性药理学实验表明西洛他唑灌流液直接作用于大鼠右心室肌细胞,可抑制Ito离子通道电流,提示西洛他唑可能对室性心律失常有影响。Objective To explore cilostazol effects on transient outward potassium current and L-Type calcium current in isolated rat right ventricular myocytes for explaining their potential anti-arrhythmic mechanism.Methods Male SD rats were randomly divided into two groups: control group and cilostazol group. The two groups’ rats were fed for 3~4 weeks, respectively. Then single ventricular myocytes were isolated from the right ventricles of SD rats by collagenase Ⅱ. Then Ito ionic current and ICa-L ionic current of two groups were recorded by using the whole cell patch clamp technique.Results In the acute pharmacological test, cilostazol of 50 μmol·L -1 and 2 μmol·L -1 significantly decreased Ito. The peak amplitude of Ito decreased from (21.55±2.61) pA/pF to (5.36±2.16) pA/pF, and (18.64±7.89) pA/pF to (7.63±1.78) pA/pF, respectively, P 〈0.05. Ito has different degrees of recovery after drug elution. There are no difference to compare with Ito and ICa-L between control group and cilostazol group ( P 〉 0.05).Conclusion Acute pharmacological experiments demonstrated that cilostazol perfused directly on rat right ventricular myocytes could inhibit Ito ion channel current. It suggests that cilostazol may have effect to ventricular arrhythmia.
