摘 要:目的通过观察急性百草枯中毒大鼠肾组织中Bcl-2、Bax、Caspase-3的表达及和TUNEL检测细胞凋亡率的变化,探索血必净对百草枯中毒的治疗作用。方法将126只Wister大鼠采用随机数字表法分为对照组、染毒组、治疗组。每组在1 d、3 d、7 d、14 d、21 d、28 d、35 d各取6只,用水合氯醛腹腔注射进行麻醉,留取肾组织标本,行免疫组化检测凋亡蛋白Bcl-2、Bax、Caspase-3的表达及TUNEL检测凋亡细胞率。结果在染毒组,大鼠肾组织中Bax、Caspase-3含量及TUNEL细胞凋亡率明显高于对照组(P<0.05),Bcl-2低于对照组(P<0.05)。经血必净治疗后,Bax、Caspase-3含量及TUNEL细胞凋亡率与中毒组比较,明显降低(P<0.05),Bcl-2在各时间点高于中毒组(P<0.05);与对照组比较,Bax、Caspase-3含量及TUNEL细胞凋亡率升高,Bcl-2含量各时段仍较低,差异有统计学意义(P<0.05)。结论 Bcl-2、Bax、Caspase-3在百草枯所致大鼠肾损伤中起重要作用;血必净增强了Bcl-2的表达,抑制了染毒大鼠肾组织中Bax、Caspase-3的含量,降低细胞凋亡率,减轻染毒大鼠肾组织损伤。Objective To investigate the therapeutic effects of Xuebijing on paraquat poisoning through observing the expressions of Bcl-2,Bax and Caspase-3 and the changes of apoptosis rate detected by TUNEL in kidney of acute paraquat poisoning rats. Methods A total of 126 Wister rats were randomly divided into control group( n = 42),contamination group( n = 42),treatment group( n = 42). At 1 d,3 d,7 d,14 d,21 d,28 d and 35 d after treatment,the 6 rats in each group were sacrificed by chloral hydrate intraperitoneal injection anesthesia. The renal tissue samples were collected and the expression of Bcl-2,Bax and Caspase-3 were detected by immunohistochemistry as well as cell apoptosis were detected by TUNEL. Results The Bcl-2,Bax and Caspase-3 content and apoptosis rate in contamination group were significantly higher than those in control group( P〈0. 05),however the expression levels of Bcl-2 in contamination group were significantly lower than those in control group( P〈0. 05). After Xuebijing treatment,the contents of Bax,Caspase-3 and the rate of apoptosis in treatment group were significantly lower than those in contamination group( P〈0. 05),and the expression levels of Bcl-2 in treatment group were significantly higher than those in contamination group in every time point( P〈0. 05). The contents of Bax,Caspase-3 and the rate of apoptosis in treatment group were significantly increased,as compared with those in control group( P〈0. 05). Moreover the content of Bcl-2 in treatment group was significantly lower than that in control group( P〈0. 05).Conclusion The Bcl-2,Bax and Caspase-3 proteins play an important role in renal injury of rats caused by paraquat poisoning. Xuebijing can enhance the expression of Bcl-2,inhibit the contents of Bax and Caspase-3 in renal tissue,decrease apoptosis rate so as to relieve the damage of renal tissue of rats with poisoning.
