摘 要:为获取更多榧属(Torreya)植物叶绿体基因组数据并开发叶绿体通用微卫星引物,对长叶榧(T.jackii)和榧树(T.grandis)进行叶绿体全基因组测序,并与已公布的的巴山榧(T.fargesii)进行比较,同时应用生物信息学软件对三者的叶绿体微卫星分布特征和规律进行全面分析。结果表明,长叶榧、榧树和巴山榧叶绿体基因组均为环状双链DNA,总长度在136720~137075bp之间。3种榧属植物的叶绿体微卫星数量以榧树中最多,为42个,其次为长叶榧(36个),巴山榧中最少(34个)。3种榧属植物叶绿体微卫星重复类型均以单碱基重复最多,其次为四碱基重复。3种榧属植物单碱基重复类型均以(A)n和(T)n为主,重复次数分别以10次和11次最多。该结果可为后续开发榧属植物叶绿体通用微卫星引物提供重要信息。In order to obtain more chloroplast genome data and develop the universal chloroplast microsatellites(cpSSR)primers for Torreya plants,chloroplast genome sequencing was performed on T.jackii and T.grandis,and compared with the published results of T.fargesii,while comprehensive analysis was conducted on the chloroplast microsatellite distribution characteristics and regular patterns of the three species by using bioinformatics software.The results showed that the chloroplast genomes of T.jackii,T.grandis and T.fargesii were circular double-stranded DNAs,and the total length was between 136720~137075 bp.The number of chloroplast microsatellites of the three Torreyaplants were the highest with 42,followed by T.jackii with 36 and T.fargesii with34.Among the repeat types of the three Torreyaplants,monocleotides was the most abundant,and the second was the Tetranucleotides.The monocleotide repeat types of the three Torreyaplants were dominated for(A)n and(T)n,and the most repeated number were 10 times and 11 times.The results provided important information for further development of universal chloroplast microsatellite primers for Torreyaplants.
