摘  要:目的探索钙调磷酸酶B同源蛋白2(CHP2)表达降低对静脉血管内皮细胞增殖和迁移的影响及其可能机制。方法通过使用靶向CHP2的si RNA降低CHP2在人脐静脉血管内皮细胞(HUVEC)中的表达,CCK-8法和流式细胞仪检测CHP2表达降低对细胞增殖和细胞周期的影响,划痕实验和Transwell实验检测CHP2对细胞迁移的作用,Western blot法检测CHP2敲降对活化T细胞核因子(NFAT)入核的影响。结果 CHP2表达降低有效抑制HUVEC细胞增殖,与G0/G1细胞周期停滞相关,CHP2敲降也抑制了HUVEC细胞迁移,机制研究表明CHP2表达降低对细胞增殖和迁移的影响可能与其抑制NFAT入核相关。结论 CHP2的表达降低抑制了静脉血管内皮细胞增殖和迁移,这可能导致静脉血管缺损修复的抑制。Objective To explore the effect of reduced expression of calcineurin B homologous protein 2 (CliP2) on proliferation and migration of venous vascular endothelial cells and corresponding mechanism. Methods CliP2 expression in human umbilical vein endothelial cells (HUVEC) was decreased by using small interfering RNA (siRNA) targeting CliP2. CCK-8 assay and flow cytometry were adopted to detect the impact of CHP2 knockdown on cell proliferation and cell cycle of HUVEC. The effect of decreased CliP2 expression on cell migration was evaluated by Scratch test and a Transwell test. In addition, the influence of CliP2 knockdown on nuclear factor of activated T cells (NFAT) nuclear translocation was detected by western blotting assay. Results CHP2 knockdown effectively inhibited the proliferation of HUVEC cells and which was related to the cell cycle arrest at G0/G1 phase. In addition, CHP2 knockdown also inhibited the migration of HUVEC cells. The mechanism study shows that the effect of CliP2 downregulation on cell proliferation and migration may be related to the inhibition of NFAT nuclear translocation. Conclusion The decreased expression of CliP2 inhibits the proliferation and migration of venous endothelial cells, which may lead to repair inhibition of damaged venous vascular.
