摘  要:目的研究利心冲剂改善心力衰竭大鼠能量代谢的作用机制。方法建立大鼠心力衰竭模型,采用利心冲剂进行干预实验,将心力衰竭大鼠分为假手术组、模型组、利心冲剂低剂量组及利心冲剂高剂量组,造模成功后将SD大鼠随机分为4组,每组10只。假手术组、模型组给予等体积双蒸水;利心冲剂低剂量组给予利心冲剂20 g/kg;利心冲剂高剂量组给予利心冲剂40 g/kg。各组大鼠灌胃给药,每日1次。给药3个月后采用Elisa法测定心肌细胞中磷酸肌酸(PCr)含量变化;Western-Blot法检测心肌组织沉默信息调节因子2同源蛋白(Sirt1)及过氧化物酶体增殖物激活受体γ辅激活因子-1α(PGC-1α)蛋白表达。结果与假手术组比较,模型组、利心冲剂低剂量组及利心冲剂高剂量组PCr含量均显著降低(P <0.05);与模型组比较,假手术组利心冲剂高剂量组PCr明显升高(P <0.05)。与假手术组比较,模型组、利心冲剂治疗组Sirt1蛋白表达明显降低(P <0.01);与模型组比较,利心冲剂低剂量组与高剂量组Sirt1蛋白表达明显增多(P <0.05或P <0.01)。与假手术组比较,模型组、利心冲剂治疗组中PGC-1α蛋白表达明显降低(P <0.01);与模型组比较,利心冲剂组PGC-1α水平有不同程度的升高,差异有统计学意义(P <0.05或P <0.01)。结论利心冲剂可改善心肌三磷酸腺苷(ATP)的传输以改善心肌能量代谢,其作用机制与增加Sirt1的活性进而激活PGC-1α介导的线粒体能量代谢途径相关。Objective To investigate the mechanism of Lixin granules (LXG) improving myocardial energy metabolism in rats with heartfailure (HF). Methods The rat model with HF was established.After successful modeling,Sprague Dawly (SD) rats were randomlydivided into 4 groups:sham group,model group,LXG high dose group (40 g/kg),and LXG low dose group (20 g/kg),10 in each group.After 3 months,the content of phosphocreatine (PCr) in cardiomyocytes was determined by enzyme linked immunosorbent assay.Theprotein expression of silent information regulator 2 homologue 1 (Sirt1) and peroxisome proliferator activated receptor γcoactivator1α(PGC 1 α ) were detected by western blot method. Results Compared with sham group,the PCr levels decreased significantly inmodel group,LXG high dose group,and LXG low dose group(P 〈0.05).Compared with model group,the PCr levels increased signif -icantly in LXG high dose group(P 〈0.05).Compared with sham group,the expression of Sirt1 protein decreased significantly in mod -el group,LXG high dose group,and LXG low dose group(P 〈0.05).Compared with model group,the expression of Sirt1 protein in -creased significantly in LXG high dose group and LXG low dose group (P 〈 0.05 or P 〈0.01).Compared with sham group,the ex -pression of PGC 1 αprotein decreased significantly in model group,LXG high dose group and LXG low dose group( P 〈0.05).Compared with model group,the expression of PGC 1 αprotein increased significantly in LXG high dose group and LXG low dosegroup (P 〈 0.05 or P 〈0.01).Conclusion LXG can improve myocardial adenosine triphosphate (ATP) transmission to improve myo -cardial energy metabolism,which is related to the increase of Sirt1 activity and the activation of PGC 1 α mediated mitochondrial en -ergy metabolism pathway.
