摘 要:目的制备载不同生长因子的复合支架及支架中释放的骨形态发生蛋白-2(BMP-2)和血管内皮细胞生长因子(VEGF)对大鼠骨髓间充质干细胞成骨分化的诱导效应。方法乳化法制备载BMP-2微球,超临界二氧化碳发泡技术制备载BMPl2微球和VEGF的复合支架,考察支架中生长因子的释放和成骨诱导作用。结果微球表面光滑,平均粒径10.022μm,包封率为64.881%,孔隙率为78.706%,能够次第释放生长因子。复合支架组细胞的增殖吸光度(A)值为(1.121±0.110)明显高于空白组(0.762±0.150)和游离生长因子组(0.882±0.140,P=0.005)。复合支架组细胞碱性磷酸酶和钙结节A值分别为(9.720±0.330、0.269±0.000),明显高于空白组[(6.242±0.660)、(0.149±0.020)%]和游离生长因子组(7.989±0.420、0.204±0.010,P=0.006、0.003)。结论支架性能良好,能够次第释放生长因子并维持其生物活性,有效地促进骨髓间充质干细胞的成骨分化。Objective To prepare scaffolds containing different growth factors, investigate the in- ductive effects of bone morphogenetic protein (BMP- 2) and vascular endothelial growth factor (VEGF) on differentiation of bone mesenchymal stem cells (BMSCs). Methods BMP - 2 - Poly D, L - Lactic Acid (PLLA) microspheres were prepared with a double emulsion method. Scaffolds were prepared with supercritical CO2 foaming technique. The effects of BMP -2 and VEGF on differentiation of BMSCs were investigated. Results BMP -2 - PLLA mierospheres were spherical shape with diameters of 10. 022 p^m. The encapsulation efficiency was 64. 881%. Scaffolds were obtained with the porosity of 78. 706% and se- quentially released rate. Cell proliferation of the scaffolds group (1. 121 +0. 110) was significantly higher than that of blank group ( 0. 762 ± 0. 150 ) and the free growth factors group ( 0. 882 ± 0. 140 ) ( P = 0. 005). ALP production and calcium nodule numbers of the scaffolds group ( 9. 720± 0. 330, 0. 269±0. 000) were significantly higher than that of blank group (6. 242±0. 660, 0. 149±0. 020) and the free growth factors group (7. 989 ± 0. 420, O. 204 ± 0. 010) ( P = 0. 006, P = 0. 003 ). Conclusion The com- posite scaffolds had excellent performance, realized the sequential delivery of BMP -2 and VEGF, and could induce BMSCs osteogenic differentiation effectively.
