一株基因10型Ⅰ类新城疫病毒P基因的遗传进化分析

摘  要:为探讨新城疫病毒(NDV)P基因及其编码蛋白的遗传进化特性,了解野鸟源NDV的基因变异情况,本研究对我国首次分离的野鸟源Ⅰ类NDV分离株SD18/13的P基因进行了测序,并从Gen Bank中选取不同基因型毒株的P基因序列进行遗传进化分析。结果表明,SD18/13 P基因开放阅读框全长1 200 bp,编码399个氨基酸,与Ⅰ类NDV的氨基酸同源性为83.5%~98.5%,与Ⅱ类NDV的氨基酸同源性为67.2%~70.5%。SD18/13与法国分离株Teal/France/10010/2010聚于一簇,同属于基因10型Ⅰ类NDV,两者P基因的核苷酸同源性为98.2%,氨基酸同源性为98.5%。In order to investigate the genetic evolution characteristics and its encoded protein of phosphoprotein( P) gene of Newcastle disease virus( NDV),and to understand the genetic variation of NDV in wild birds,the P gene of SD18/13 firstly isolated from wild type bird NDV in China was sequenced. The P gene sequences of different genotypes were selected from Gen Bank,and genetic evolution analysis was carried out.The results showed that P gene of SD18/13 was 1 200 bp in length and contained an opening reading frame coding 399 amino acids. P gene of SD18/13 share 83. 5% - 98. 5% amino acid homology with Class ⅠNDV,and 67. 2% - 70. 5% amino acid homology with Class Ⅱ NDV. SD18/13 was clustered into sub-genotype 10 in Class Ⅰ NDV,and showed high similarity with French isolate Teal/France/10010/2010. The nucleotide homology of the P gene between SD18/13 and Teal/France/10010/2010 was 98. 2% and the amino acid homology was 98. 5%.
