摘 要:桅子是一种具有重要药理作用的常用传统中药,并被卫计委列为首批既是食品又是药品中药名录。桅子中黄色素含量较高,水溶性好,无毒性,国内外已将其广泛应用为食品添加剂和工业染料,但目前桅子黄生产工艺复杂,产品得率和色价均比较低。为综合开发高品质的栀子黄色素,本研究采用单因素及正交试验确定了栀子黄色素的最佳浸提条件:提取时间3 h,提取温度50℃,料液比1∶8;通过考察5种不同类型大孔树脂对桅子黄色素的静态吸附性能,最终确定HP2MGL为最佳精制材料,采用梯度洗脱法定量洗脱,最终产品色价由最初的68.79增至340.00,得率为2.2%,达到了精制的目的。该研究为工业化提取和精制桅子黄色素提供了理论基础。Gardenia is a commonly used traditional Chinese medicine with important pharmacological functions,and it is listed as food and medicine by the National Health Commission of China. The yellow pigment in gardenia fruit has a high content,good water solubility and non-toxic,and has been widely used as a food additive and an industrial dye in the world. But at present,the production process of gardenia pigment is complex,and its yield and color value are both very low. In order to develop high-quality gardenia yellow pigment,the extraction conditions were optimized by combination of single-factor test and orthogonal test as extraction time of 3 hours,extraction temperature at 70℃ and material to liquid ratio of 1∶8. HP2 MGL was finally selected as the material for refining gardenia yellow pigment after the evaluation on the static adsorption property to yellow pigment of 5 types of macroporous resin. The gradient elution method was adopted to elutegardenia yellow pigment and the color value of refined gardenia yellow pigment increased from 68. 79 to 340. 00 with a high quality. The study provided a theoretical basis for industrial extraction and refining of yellow pigment from gardenia.
