摘 要:目的:本研究通过观察真武汤对转基因扩张型心肌病(DCM)小鼠心肌组织Atp2a2、Col1a1基因及蛋白表达变化,探究真武汤防治DCM的分子生物学机制。方法:将cTnT^(R141W)转基因DCM小鼠60只随机分为模型对照组,西药组,真武汤高、中剂量组,C57BL/6J小鼠15只作为空白对照组,疗程4周。采用q RT-PCR法检测各组心肌组织Atp2a2、Col1a1基因表达;Western Blot法检测各组心肌组织Atp2a2、Col1a1蛋白表达。结果:与空白对照组比较,模型对照组Atp2a2 mRNA及蛋白表达显著下降(P<0.01),Col1a1 mRNA及蛋白表达显著升高(P<0.01),与模型对照组比较,真武汤各剂量组Atp2a2 mRNA表达均不同程度上调(P<0.05,P<0.01),Col1a1 mRNA及蛋白表达不同程度下调(P<0.05,P<0.01),真武汤高剂量组上调Atp2a2,下调Col1a1 mRNA及蛋白表达作用最佳。结论:真武汤通过上调心肌组织中Atp2a2 mRNA及蛋白表达,下调Col1a1 mRNA及蛋白表达,从而抑制心肌细胞肥大及纤维化过程,起到防治DCM的作用。Objective: To observe the changes of Atp2 a2 and Col1 a1 gene and protein expression in myocardium of transgenic mice with dilated cardiomyopathy(DCM), and explore the molecular mechanism of Zhenwu Decoction in the prevention and treatment of dilated cardiomyopathy. Methods: The cTnT^(R141W) gene was transfected into DCM mice to establish a transgenic mouse model of cTnT^(R141W) gene specific for myocardium, fifteen C57 BL/6 J mice were used as blank control group, 60 mice in the model group were randomly divided into model control group, western medicine group, high and middle dose Zhengwu Decoction group. After 4 weeks, the mRNA expression of Atp2 a2 and Col1 a1 in myocardium were detected by q RTPCR. The expression of Atp2 a2 and Col1 a1 protein was detected by Western Blot. Results: Compared with the control group, the expression of Atp2 a2 mRNA and protein was significantly decreased(P〈0.01), the expression of Col1 a1 mRNA and protein was significantly increased(P〈0.01), and the expression of Atp2 a2 mRNA in the model control group was significantly higher(P〈0.01). The expression of Col1 a1 in the high and middle dose Zhengwu Decoction group were significantly higher than that of the model control group(P〈0.05, P〈0.01), and the mRNA and protein expression of Col1 a1 was significantly decreased(P〈0.05, P〈0.01), and the high dose Zhengwu Decoction could better raise Atp2 a2, and better downregulation Col1 a1 mRNA and protein expression. Conclusion: Zhenwu Decoction could increase the mRNA and protein expression of Atp2 a2 and decrease the mRNA and protein expression of Col1 a1 in myocardium to inhibit the hypertrophy and fibrosis of cardiomyocytes, then prevent the attack of DCM.
