摘  要:目的:研究知柏地黄汤对解脲脲原体(UU)感染模型大鼠精子线粒体膜电位(MMP)、活性氧(ROS)水平及细胞色素C(CytC)含量的影响。方法:雄性SD大鼠80只,随机分成假手术组、模型组、中药组(知柏地黄汤,2mL/d)、中西组(知柏地黄汤+强力霉素)、西药组(强力霉素,2mL/d),除假手术组外,其他组均采用经大鼠膀胱注射UU建立UU感染动物模型。连续灌胃给药21d后处死取标本检测,采用荧光流式细胞技术(JC-1染色)测定精子MMP水平(JC-1+%)及ROS水平(JC-1+%),采用ELISA法检测CytC含量。结果:模型组大鼠精子MMP(JC-1+%)、ROS水平(JC-1+%)及精子中CytC含量,与假手术组比较,差异显著(P<0.01);各治疗组中精子MMP水平上升,ROS水平、CytC含量下降,其中,中西组与模型组比较,差异显著(P<0.01)。结论:知柏地黄汤能够提高UU感染大鼠精子MMP水平,减少精子ROS产生,减少精子凋亡,抑制CytC释放,这可能是知柏地黄汤治疗UU感染所致男性不育症的机制之一。Objective: To study the effect of Zhibai Dihuang Decoction(ZDD) on the mitochondrial membrane potential(MMP), reactive oxygen species(ROS) level and cytochrome C(CytC) content in the sperm of ureaplasma urealyticum(UU) infection model rats. Methods: A total of 80 male SD rats were randomly divided into the sham operation group, the model group, the Chinese materia medica group(ZDD, 2 mL/d), the combined Chinese materia medica and western medicine group(ZDD+Doxycycline), and western medicine group(Doxycycline, 2 mL/d). In addition to the sham operation group, UU infection models were established by injection of UU in rats. After intragastric administration of 21 d, the samples were sacrificed. The levels of sperm MMP(JC-1+%) and ROS(JC-1+%) were determined by fluorescence flow cytometry(JC-1 staining), and the content of CytC was detected by ELISA. Results: The level of MMP, ROS and the content of CytC of sperm in the model group rats and the sham operation group were different. There was a significant difference between these two groups(P〈0.01). The level of MMP, ROS and the content of CytC in the treatment group were decreased. Compared with the UU infection model group, the difference was significant(P〈0.01). Conclusion: ZDD can increase the MMP level of sperm in UU infected rats, reduce ROS production, reduce sperm apoptosis and inhibit CytC release. This may be one of the mechanisms of ZDD in the treatment of infertility caused by UU infection.
