摘  要:目的:运用逍遥散加减方干预高催乳素血症(HPRL)模型大鼠并探讨其对卵巢颗粒细胞(GC)凋亡的影响及相关机制。方法:将48只SD雌性性成熟大鼠随机分为空白组,模型组,中药高、中、低剂量组,溴隐亭组。选用腹腔注射甲氧氯普胺制作HPRL动物模型,采用免疫组化检测各组中Bcl-2、Bax的含量;蛋白印迹法检测各组中细胞色素C(CytC)、Caspase-3、Caspase-8和Caspase-12的含量;运用TUNEL法检测GC凋亡阳性细胞数。结果:与空白组比较,模型组GC中Bcl-2低表达,Bax、CytC、Caspase-8、Caspase-12、Caspase-3、凋亡小体高表达(P<0.01),而与模型组比较,中药高剂量组GC中Bcl-2高表达,Bax、CytC、Caspase-8、Caspase-12、Caspase-3、凋亡小体低表达(P<0.01,P<0.05)。对比中药各剂量组间关系发现,其存在一定的量效关系。结论:逍遥散加减方可通过降低Bax、CytC、Caspase-3、Caspasw-8和Caspase-12含量,提升Bcl-2的含量进而下调GC凋亡阳性数目,且和中药成一定正比量效关系,这可能是其改善卵巢功能的机制之一。Objective: To investigate the effect and mechanism of the apoptosis of Xiaoyao Powder by using the free and loose addition and subtraction. Methods: A total of 48 SD rats were randomly divided into blank group, modeling group, high, medium and low dose group, and positive group. The HPRL animal model was prepared by ip. Methoxyclopamine. The contents of Bcl-2 and Bax in each group were detected by immunohistochemistry. The contents of CytC, Caspase-3, Caspase-8 and Caspase-12 in each group were detected by the protein imprinting method. The number of positive cells of ovarian granule cell apoptosis was detected by TUNEL method. Results: Compared with the blank group, the content of Bcl-2 in modeling group was significantly lower in the formation of GC, the contents of Bax, CytC, Caspase-3, Caspase-8 and Caspase-12 and apoptotic body were increased(P〈0.01), however, compared with the modeling group, the high close group each indexes were contrary(P〈0.01, P〈0.05). It was found that the dosage groups showed a certain amount of effect. Conclusion: Xiaoyao Powder could through reduce Bax, CytC, Caspase-3, Caspase-8 and Caspase-12 levels, improve the content of the Bcl-2 and then cut ovarian GC apoptosis positive number, and traditional Chinese medicine is a certain effect than quantity relationship, this could be one of the mechanism of improve ovarian function.
