葛根芩连汤治疗溃疡性结肠炎谱效关系研究

摘　　要：目的:研究葛根芩连汤治疗溃疡性结肠炎谱效相关性,探讨葛根芩连汤药效物质基础。方法:在建立葛根芩连汤UPLC-MS/MS指纹图谱和获得葛根芩连汤对湿热内蕴型溃疡性结肠炎大鼠模型血清及组织液TNF-α影响的药效信息基础上,采用主成分分析及灰色关联度分析将葛根芩连汤主要化学成分与药效信息相关联。结果:获得葛根芩连汤的指纹数据和药效信息,葛根芩连汤UPLC-MS/MS指纹图谱的主要共有峰与其药效作用的关联度均>0.5。葛根芩连汤发挥药效作用是葛根素、大豆苷、黄连碱、表小檗碱、盐酸药根碱、黄芩苷、盐酸小檗碱、盐酸巴马汀、汉黄芩苷、黄芩素、甘草酸铵等多成分。结论:通过谱效关系研究确定了葛根芩连汤发挥药效作用是其主要成分共同作用的结果,为其质量评价与控制、药效物质基础研究提供依据。Objective： To study the spectrum-effect relationship and probe the pharmacodynamic material basis of Gegen Qinlian Decoction. Methods： Based on the establishment of UPLC-MS/MS fingerprint of Gegen Qinlian Decoction and obtainment of pharmacodynamic information of its effect on TNF-α contents of rat＇s serum and interstitial fluid of the dampheat ulcerative colitis, the main chemical constituents of Gegen Qinlian Decoction were correlated with the pharmacodynamic information by grey correlation degree analysis. Results： The fingerprint data and efficacious information of Gegen Qinlian Decoction were obtained, and the correlation degree between the common pattern of UPLC-MS/MS fingerprint of Gegen Qinlian Decoction and its effect all reached 0.5. The efficacy of Gegen Qinlian Decoction was due to the combination of multicomponent of puerarin, daidzin, coptisine, epiberberine, jatrorrhizine, baicalin, berberine, palmatine, wogonoside, baicalein, ammonium glycyrrhizinate. Conclusion： According to the research on spectrum-effect relationship, the result indicates that the efficacy of Gegen Qinlian Decoction is due to the combination of multicompnnent, it provides the basis for quality evaluation and control, and effective substance base research.
