摘 要:目的:探讨糖耐量低减(IGT)大鼠血管内皮炎性反应机理及中药复方加减参麦汤的调控机制。方法:采用二联法建立IGT大鼠模型。采用ELISA法、HE染色法、Envision免疫组化染色法分别检测C反应蛋白(CRP)、内皮素-1(ET-1)、细胞间黏附分子-1(ICAM-1)、血管细胞黏附分子-1(VCAM-1)水平、主动脉形态变化及主动脉VCAM-1蛋白表达。结果:模型组大鼠与正常组比较,CRP、ET-1、ICAM-1及VCAM-1水平升高明显(P<0.01),加减参麦汤组与模型组比较,上述指标显著降低(P<0.01)。模型组大鼠血管内皮黏附较多单核细胞,细胞体积增大,间隙增大,内膜不光滑,血管壁增厚,内膜处可见泡沫细胞,少量单核及巨噬细胞浸润,加减参麦汤组病理变化较模型组相对改善。模型组大鼠动脉血管VCAM-1表达较正常组明显增强(P<0.01),加减参麦汤组可改善模型组大鼠VCAM-1在主动脉的表达(P<0.01)。结论:IGT大鼠存在血管内皮炎性反应。中药复方加减参麦汤对IGT大鼠的血管内皮炎性反应具有保护作用,其降糖作用与二甲双胍类似。Objective: To explore the mechanism of vascular endothelial inflammatory response and the regulation and control mechanism of modified Shenmai Decoction in impaired glucose tolerance (IGT) rats. Methods: The model of IGT rats was established by the two-link method. The levels of CRP, ET-1, ICAM-1 and VCAM-1 were detected by ELISA method. HE staining was used to detect aortic morphological changes, The VCAM-I protein expression of aorta was detected by the Envision cal staining method. Results: The levels of CRP, ET-1, ICAM-1 and VCAM-1 were significantly increased in IGT rats compared with the normal group (P〈0.01), and the above indexes were significantly decreased in the modified Shenmai Decoction group compared with the model group (P〈0.01). There were some changes in model group, such as more mononuclear cells in rat's vascular endothelial adhesion, increased cell volume, increased gap, not smooth intima, thickening of the vessel wall, foam cells in intima visible, a few mononuclear and macrophage infiltration, and compared with model group, the pathological changes of TCM treatment group were relatively improved. The expression of VCAM-1 in rats arterial vessels was significantly enhanced (P〈0.01), The expression of VCAM-1 in the aorta was improved by modified Shenmai Decoction (P〈0.01). Conclusion: IGT rats have endothelial inflammatory response. Modified Shenmai Decoction has a protective effect in IGT rats' vascular endothelial inflammatory response, and the hypoglycemic effect of modified Shenmai Decoction is similar to metformin.
