摘 要:目的:从附子黑顺片中提取粗多糖,并对其体内外免疫活性进行研究。方法:采用水提醇沉的方法提取黑顺片中粗多糖,苯酚-硫酸法测定其多糖含量,气相色谱法分析其单糖组成;分别用不同剂量的黑顺片多糖(200、100、50mg·kg^(-1)·d^(-1))灌胃由环磷酰胺所致的免疫低下模型小鼠,灌胃7d后检测脏器指数及一氧化氮(NO)、干扰素γ(IFN-γ)等指标;用MTT法研究黑顺片多糖对小鼠脾脏细胞、腹腔巨噬细胞的免疫作用影响。结果:黑顺片多糖含量为1.78%,其单糖组成主要是D-阿拉伯糖、D-木糖、D-葡萄糖和D-半乳糖;黑顺片多糖溶液均可促进脾脏和胸腺指数,高剂量组尤为明显(P<0.01);黑顺片多糖浓度为25μg/mL和25~100μg/mL时可分别促进小鼠脾脏细胞和腹腔巨噬细胞的增殖(P<0.05,P<0.01);高剂量组小鼠血清中NO和IFN-γ含量较模型组显著升高(P<0.05,P<0.01)。结论:黑顺片多糖对固有免疫及适应性免疫均有一定增强作用。Abstract: Objective: To extract polysaccharide from Heishunpian, and to primarily study on its immune activity. Methods: Polysaccharide was extracted from Heishunpian by water-extraction-alcohol-precipitation method, and determined by phenol sulfonic acid spectrophotometry. The monosaccharide composition was analyzed by gas chromatography. The mice were treated with different doses of Polysaccharide (200, 100, 50mg ·kg^-1. d^-1) for 7 days after intragastric administration of cyclophosphamide. The organ index, NO and IFN- 7 content were measured. The effects of polysaccharide on the immune function of mouse spleen cells and peritoneal macrophages were detected by MTT assay. Results: The content of polysaccharide of samples was 1.78%, and the polysaccharide was composed of D-arabinose, D-Xylose, D-glucose and D-galactose. Polysaccharide from Heishunpian could improve the spleen and thymus index, especially in high-dose group (P〈0.01). The proliferation of spleen ceils and peritoneal macrophages in mice were promoted when the concentration of polysaccharides was 25μg/mL and 25-100μg/mL (P〈0.05, P〈0.01). NO and IFN-γ level in serum from high-dose group increased significantly (P〈0.05, P〈0.01). Conclusion: Polysaccharide from Heishunpian could significantly improve the innate and adoptive immunity in mice.
