摘  要:目的:研究清脑滴丸对脑缺血再灌注大鼠海马Janus激酶2/信号转导子和转录激活子3(JAK2/STAT3)磷酸化蛋白(p-JAK2/p-STAT3)表达的影响。方法:采用线栓法制备大鼠大脑中动脉缺血(MCAO)再灌注模型,分为假手术组、MCAO组、清脑滴丸组。MCAO 1.5h再灌注24h处死大鼠,Garcia J H评分检测神经功能缺损程度,TTC染色检测脑梗死体积,Western blot法检测各组缺血侧海马JAK2/STAT3及其磷酸化蛋白p-JAK2/p-STAT3表达。结果:MCAO组脑梗死体积增大;与假手术组比较,MCAO组神经功能评分降低(P<0.05),p-JAK2/JAK2,p-STAT3/STAT3蛋白表达明显升高(P<0.01);与MCAO组比较,清脑滴丸组神经功能评分升高且脑梗死体积降低(P<0.05),p-JAK2/JAK2,p-STAT3/STAT3蛋白表达显著升高(P<0.01)。结论:清脑滴丸可能通过激活JAK2/STAT3磷酸化,发挥对脑缺血再灌注的神经保护作用。To study the effects of Qingnao Dripping Pills(QNDP) on the protein expression of Janus kinase 2/signal transducer and activator of transcription 3(JAK2/STAT3) phosphorylation(p-JAK2/p-STAT3) in hippocampus of rats with cerebral ischemia-reperfusion. Methods: The rat models of focal cerebral ischemia and reperfusion were made by thread occlusion technique induced middle cerebral artery occlusion. Rats were randomly divided into 3 groups: sham group, MCAO group and QNDP group. Rats were sacrificed after 1.5 h MCAO and 24 h reperfusion. Neurological deficits were evaluated by Garcia JH neurological score, cerebral infarct volume were measured by TTC staining. The protein expression of p-JAK2, JAK2, p-STAT3, STAT3 in ischemic hippocampus were detected by Western blot. Results: Cerebral infarction volume of the MCAO group was increased. Compared with the sham group, the neurological score of the MCAO group was decreased(P〈0.05), the protein expression of p-JAK2/JAK2 and p-STAT3/STAT3 were significantly increased in the MCAO group(P〈0.01). Compared with the MCAO group, the neurological score of the QNDP group was increased and cerebral infarction volume was decreased(P〈0.05), the protein expression of p-JAK2/JAK2 and p-STAT3/STAT3 were significantly increased(P〈0.01). Conclusion: The protective effects of QNDP against cerebral ischemia-reperfusion may be related to activate JAK2/STAT3 phosphorylation.
