摘 要:选择常用的多平台水环境法和睡眠剥夺箱两种方法进行睡眠剥夺,观测动物行为表现,探究不同睡眠剥夺对小鼠情绪和学习记忆的影响。结果显示,12、24h短期睡眠剥夺,两种剥夺方法均不会对小鼠的情绪和学习记忆产生影响。48h的睡眠剥夺、睡眠剥夺箱并没有造成小鼠焦虑样情绪和空间记忆能力受损,但多平台水环境法会使小鼠出现焦虑样情绪,空间记忆未受影响。用睡眠剥夺箱进行长时间睡眠剥夺,小鼠均产生焦虑样情绪;剥夺时间达到3周,学习过程减慢,空间记忆能力受损。睡眠剥夺箱在长期睡眠剥夺研究中可成功建立模型,能够模拟人类长期睡眠不足状态。To investigate the effect of different sleep deprivations(SD)on the mood,learning and memory of mice,both multiple platform method and sleep deprivation tank method were used to observe the effects of the relative factors on the anxiety-like behavior,learning and memory.The following results were obtained.12 hSD and 24 hSD did not affect anxiety-like behavior,learning and memory in either of the above paradigms.The anxiety-like behavior was observed in mice subjected to 48 hSD via multiple platform method and the spatial memory was not affected,but sleep deprivation tank did not make mice appear anxiety-like behavior and spatial memory ability after 48 hSD.In the sleep deprivation tank experiment,a long-time SD could induce anxiety-like behavior,whereas the learning process of the mice slowed down and spatial memory was impaired after 3 weeks SD.Thus,deprivation tank is advantageous as an SD model for studies on human long-term sleep insufficiency.
