摘 要:本文旨在探讨白纹伊蚊Aedes albopictus唾液腺匀浆粗蛋白(salivary gland extract,SGE)血小板聚集抑制和抗凝血活性,为后续单个唾液蛋白在蚊虫吸血中的功能研究奠定基础。采用比浊法检测低、中、高浓度SGE在不同血小板聚集激活剂作用下对人血小板聚集活性的影响,手工法检测不同浓度SGE对人全血部分凝血活酶时间,凝血酶原时间,凝血酶时间的影响。结果显示,低浓度SGE对Ⅰ型胶原诱导的血小板聚集有显著抑制作用,随着浓度升高具有明显的量效关系,最高抑制率可达90. 36%;高浓度SGE对凝血酶诱导的血小板聚集具有显著抑制作用。SGE对ADP诱导的血小板聚集无明显抑制作用。SGE对人血浆APTT、PT、TT均有延迟效应,1. 25 ng/m L SGE即对TT有显著延迟效应。研究结果表明,白纹伊蚊SGE不仅对胶原诱导的血小板聚集有明显的抑制作用,而且可作用于血液级联凝集全过程。The platelet aggregation inhibition and anti-coagulant activity were detected to evaluate the functions of salivary gland extracts (SGE) of Ae. albopietus. The effect of platelet aggregation inhibition was monitored by turbidimetry with low, medium and high concentrations of SGE when induced by type I collagen, thrombin and ADP, respectively. The platelet aggregation induced by type I collagen was inhibited obviously by low concentrate SGE ( P 〈 0.05) with significant dose- effect relationship as the concentration increased, the highest inhibition rate was 90% ; the platelet aggregation induced by thrombin was inhibited significantly by high concentrate SGE (P 〈 0. 05 ) , SGE showed no obvious inhibition effect on platelet aggregation induced by ADP. Under certain concentration of SGE, APTT, PT and TT extended significantly (P 〈 0.01 ) , and TT was extended at 1.25 ng/mL SGE ( P 〈 0.05 ). The results showed that SGE has significant inhibition activity on platelet aggregation induced by collagen and can interact at all levels of coagulation (the intrinsic, extrinsic and common pathways).
