摘 要:以海南、福建等省常见库蠓为研究对象,构建基于ITS的吸血库蠓成虫分子鉴定体系。共24种库蠓的72条序列用于比较分析、遗传距离和系统发育分析显示:24种库蠓的种间遗传距离为0. 05~0. 39,平均遗传距离0. 25;种内遗传距离为0~0. 13,平均遗传距离0. 07。种间遗传距离为种内遗传距离的3. 57倍,表明ITS基因具有较大的种间甚至种内变异性,是理想的靶标基因,可作为库蠓种类鉴定的分子标记。Specimens of biting midges collected from Hainan, Fujian, Guangxi, Heilongjiang and Xinjiang province and autonomous region were studied to construct the molecular identification system of adult Culicoides. Twenty-four species of Culicoides adults and 72 ITS gene sequences were analyzed. It was showed from genetic distance and by phylogenetic analysis that the interspecifie genetic distance was 0.05 - 0. 39 with average distance of 0.25, and the intraspecifie genetic distance was 0 - 0. 13 with average distance of 0.07. The interspecific genetic distance was 3.57 times of intraspecific genetic distance, indicating that ITS genes might be employed as one of suitable molecular markers for identifying the bloodsucking midges.
