摘 要:目的探讨蛇菰多糖(BPS)对实验性糖尿病大鼠过氧化物酶体增殖物激活受体γ(PPARγ)、鸢尾素及糖脂代谢的影响及机制。方法雄性SD大鼠饲喂高脂饲料(在普通饲料中添加猪油10%、鸡蛋5%和胆固醇2.5%)4周后,一次性ip给予链脲佐菌素(STZ)40 mg·kg-1,1周后测定空腹血糖(FBG),FBG≥7.8 mmol·L-1确认模型成功。模型大鼠ig给予BPS 200 mg·kg-1,持续4周后,处死大鼠。取肝组织冰冻切片进行苏丹Ⅲ染色,观察脂肪变性程度;检测血糖、甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)和血清鸢尾素含量及肝超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量的变化;Western蛋白印迹法测定肝组织PPARγ蛋白表达。结果与正常对照组比较,模型组大鼠体质量增加幅度降低(P<0.01),肝指数增加(P<0.01),肝细胞内可见大量脂滴,并出现了不同程度的炎症细胞浸润,血清TG,TC和LDL-C水平明显升高(P<0.01),HDL-C显著降低;肝组织SOD活性降低,MDA含量增加(P<0.01);血清鸢尾素水平降低(P<0.01);肝组织中PPARγ蛋白表达减少(P<0.01)。与模型组比较,BPS治疗组大鼠体质量增加幅度明显增加(P<0.01),大鼠肝脂肪变性和炎症程度明显改善,血清TG,和LDL-C水平明显降低,HDL-C显著升高(P<0.05);肝组织SOD活性升高,MDA含量减少(P<0.05);血清鸢尾素水平升高(P<0.05);肝组织中PPARγ蛋白表达增加(P<0.05)。结论 BPS能明显降低STZ致糖尿病大鼠的血糖水平,增强抗氧化能力,改善血脂代谢异常,提升鸢尾素水平并增强肝组织PPARγ的表达。OBJECTIVE To investigate the effects and mechanisms of Balanophora polysaccharide(BPS) on expressions of PPARγ, irisin and glucolipid metabolism in experimental diabetic rats. METHODS Male SD rats were fed with high fat feed(10% lard, 5% eggs and 2.5% cholesterol were added to normal feed) for 4 weeks, while streptozocin(STZ, 40 mg·kg^-1) was injected into the abdominal cavity at one time. Fasting blood glucose(FBG) was measured one week later. The model group was identified when the level of FBG ≥7.8 mmol·L-1. Then the model group was ig administered with BPS 200 mg·kg^-1 for 4 weeks. Liver fatty degeneration was observed by frozen sections with SudanⅢstaining. The expression of PPARγ protein in the liver was measured by Western blotting, blood glucose, triglycerides(TG),total cholesterol(TC), low density lipoprotein cholesterol(LDL-C), high-density lipoprotein cholesterol(HDL-C) and content of serum irisin were tested, and the activity of superoxide dismutase(SOD) and the level of malondialdehyde(MDA) in the liver tested. RESULTS The levels of serum TG, TC, LDL-C and hepatic index in model group were significantly increased(P〈0.01) while HDL-C level, SOD activity and irsin level were significantly decreased compared with normal control group(P〈0.01), and PPARγexpression was also downregulated(P〈0.01). The levels of TG and LDL-C decreased, SOD activity and irisin levels was increased, MDA content lowered(P〈0.05), and expression of PPARγ protein in the liver increased(P〈0.05) compared with model group. The body mass was significantly increased(P〈0.01), but fat degeneration and inflammation were significantly reduced after BPS treatment(P〈0.05). CONCLUSION BPS can significantly reduce the blood glucose level in diabetic rats induced by STZ, enhance the antioxidant capacity, improve the abnormal blood lipid metabolism, improve the irisin level and enhance the PPAR expression in the liver.
