摘  要:【目的】拟南芥是研究植物基因功能的工具材料,在图位克隆进行基因定位时面对数千个分子标记常常让新的研究者无从下手。【方法】本文通过从先前筛选的44个分子标记,进一步筛选出了24个平均分布于拟南芥五条染色体上的遗传标记。【结果】这24个标记在PCR检测中易于扩增、实验条件相差小、电泳条带区分度好,是进行图位克隆的理想分子标记。【结论】利用这些标记,建立了拟南芥初定位快速图位克隆系统,大大提高了初定位的效率,并实现拟南芥盐敏感突变体S28的快速染色体初定位。[ Ohjective]Arabidopsis thaliana was a tool for studying the function of plant genes. It was often difficult for new researchers to figure out thousands of molecular markers when mapping genes for positional cloning. [ Method] In this study, 24 genetic markers distributed on five chromosomes of Arabidopsis thaliana were screened out by using 44 molecular markers screened previously. [ Result] These 24 markers were easy to amplify in PCR detection, the experimental condition was relatively consistent, and the electrophoresis bands divisions were good, and it was an ideal molecular marker for map cloning. [ Conclusion] Using these markers, fast cloning system of the Arabidopsis was established, which greatly improved the efficiency of initial positioning, and achieved rapid initial localization of chromosome of Arabidopsis salt sensitive mutants S28, had proved that the method was efficient and practical.
