口服湿热痹胶囊大鼠体内代谢产物分析

摘  要:目的鉴定湿热痹胶囊给予大鼠后血浆、尿液和粪便中的代谢产物。方法色谱柱为Waters C18(2.1 mm×50 mm,1.7μm),柱温40℃。流动相:0.1%甲酸水溶液-0.1%甲酸乙腈溶液,梯度洗脱。超高压液相色谱-飞行时间质谱仪采用电喷雾电离离子源,正、负离子模式检测。结果超高效液相色谱-质谱联用法分析出湿热痹胶囊中7种入血的原型成分,在尿液中分析出湿热痹胶囊原型成分6种,粪便中分析出湿热痹胶囊原型成分11种。结论体内测得成分可能为湿热痹胶囊发挥药效的物质基础。Objective To identify the metabolites in rat plasma,urine and faces after oral administration of SHIREBI capsule. Methods Column of Waters C 18 (2.1 mm × 50 mm,1.7 μm) was used at 40 ℃,folic acid contained water and acetonitrile was used as mobile phase in a gradient elution.Analysis of HPLC-QTOF MS with electrospray ion source was conducted in both positive and negative mode. Results Seven,six and eleven kinds of metabolites were identified by the proposed method. Conclusion The prototype metabolites found in plasma maybe the effective compounds in Shirebi capsule.
