摘  要:通过中医传承辅助系统挖掘痔疮治疗新方,联合整合药理学平台分析新方潜在作用分子靶点和信号通路.以《中医方剂大辞典》为基础汇编痔疮治疗中药方剂数据库,通过辅助系统软件计算获得药物使用频次并挖掘新方.以整合平台提供的功能模块进行靶标推算、网络布局等计算,建立新方的成分-靶标网络、新方-痔疮相关靶标网络,在功能分析和通路分析结果推导并揭示了新方作用的痔疮相关靶标分子机制.结果表明:完成对痔疮组方数据库提供的295个方剂进行分析,计算得到方剂药物使用频次,获得3首新方剂,深入研究了新方剂的分子机制.运用中医传承辅助系统以及整合药理学平台,可以从宏观和微观角度对痔疮的中药治疗挖掘新方并研究潜在分子机制,为研究中医方剂的计算推演提供了方向指导.Based on traditional Chinese medicine inheritance system and integrated pharmacology platform for data mining of hemorrhoids prescription, we screened the difference of molecular mechanism between high-frequency herb combination and potential new prescriptions. The prescriptions of hemorrhoids were collected from Chinese traditional medicine dictionary. Association rules and complex system entropy clustering were used to determine the frequency and association rules of drug use, and to identify high-frequency herb combinations and new potential prescriptions. By using integration platform, network construction and target prediction, disease-target network, herb-target network, GO and KEGG pathway of hemorrhoids related preparations of high frequency as well as potentially new pairs were analyzed. Results show that from 295 prescriptions, the frequency of herb use and the association rules were analyzed. 3 new potential prescriptions were identified, and the disease-target relationship obtained, and molecular mechanism differences between the high-frequency herb combination and the new potential prescription were compared.In conclusion,combination of Traditional Chinese Medicine inheritance system and integrated pharmacology platform can help explore the TCM formula and potential molecular mechanism of hemorrhoids treatment macroscopically as well as microscopically.
