不同配制比例的当归/白芍提取物对原发性痛经大鼠的作用研究

摘　　要：目的:探讨当归/白芍组合物对原发性痛经大鼠的干预作用,并从不同药物配置浓度中发现其对治疗原发性痛经的最优化配比。方法:雌性未孕Wistar大鼠随机分为空白组、模型组、配比组(1∶1、1∶3、1∶5、1∶6、2∶1、2∶3)、单味当归组、单味白芍组,采用缩宫素建立原发性痛经大鼠模型,观察各组大鼠发生扭体的潜伏期及发生次数,计算扭体反应抑制率。同时检测各组大鼠子宫组织中一氧化氮(NO)及内皮素(ET)的含量。结果:与空白组比较,模型组大鼠发生扭体的潜伏期缩短,扭体次数增加,子宫组织中NO含量降低,ET含量明显增加(P<0.05)。与模型组比较,配比组2、3、4大鼠扭体潜伏期明显延长,扭体次数减少,差异具有显著性,其中配比组3大鼠扭体抑制率最高;配比组2、3、4、5、8大鼠子宫组组中NO含量明显增加(P<0.05),配比组3、4大鼠ET含量明显下降,差异具有显著性(P<0.05)。结论:当归/白芍组提取物可通过抑制子宫收缩,改善子宫供血,从而对原发性痛经产生治疗作用,其中1∶5的配置比例疗效最好。Objective：To explore the intervention effect of Angelica sinensis/Paeonia lactiflora Pall.composition on primary dysmenorrhea rats,and found that the treatment of primary dysmenorrhea from different drug concentration ratio optimization allocation.Methods：Female non pregnant Wistar rats were randomly divided into controlled group,model group,matched group（1：1,1：3,1：5,1：6,2：1,2：3）,single Angelica sinensis group,single herb Paeonia lactiflora Pall.group,the use of oxytocin to establish primary dysmenorrhea rats model,rats were observed occurrence of writhing latency and the number of times of occurrence,calculated the inhibition rate of writhing.Simultaneous detection of nitric oxide of rats in each group（NO） and endothelin（ET） content.Results：Compared with the blank group,the latent period of writhing in the model group was shortened,the number of writhing was increased,the content of NO in the uterine tissue was decreased,and the content of ET was increased significantly（P〈0.05）.Compared with the model group,the ratio of group 2,3,4 rats writhing latency was prolonged and reduced writhing times,the difference was significant,the ratio of group 3 rats writhing the highest inhibition rate;In group 2,3,4,5,8,the content of NO in the uterus group increased significantly（P〈0.05）,and the content of ET in the 3,4 group decreased significantly,and the difference was significant（P〈0.05）.Conclusion：Angelica sinensis/Paeonia lactiflora Pall.combination can inhibit uterine contraction,improve uterine blood supply,and thus have therapeutic effect on primary dysmenorrhea,in which the ratio of 1：5 configuration is the best.
