摘  要:为鉴定所分离的貉源嗜水气单胞菌(A.hydrophila)生物学特性,本试验从无菌采集的10份病死貉的肝脏、心血、肺脏、脾脏等病料组织中分离到相同的单一病原菌,将其命名为HBA-2。通过细菌的生物特性检验与16SrRNA PCR方法对HBA-2进行鉴定。16Sr RNA基因序列的系统发育树分析结果表明,HBA-2与A.hydrophila参考菌株同源性达100%,HBA-2为貉A.hydrophila。动物回归试验结果表明,HBA-2对貉和小鼠具有很强的致病性。用PCR方法检测HBA-2的4种毒力基因aer、hlyA、epa、act,结果显示,HBA-2携带hlyA、epa2种毒力基因。免疫保护性试验结果显示,HBA-2具有很好的免疫原性。药敏试验结果显示,HBA-2对头孢曲松、恩诺沙星、阿米卡星、氟苯尼考、头孢克肟5种药物高敏,对其他药物具有不同程度的耐药。One strain of pathogenic bacterium named HBA-2 was isolated from the 10 dead raccoon dog and identify for the biologic characteristics.The disease tissue were sampled from the liver,heart blood,lung,spleen,etc.HBA-2 was identified as A.hydrophila by biochemical characteristics test of bacterium and 16 SrRNA PCR method.The result of 16 SrRNA gene phylogenetic tree showed that HBA-2 shared 100% homology with A.hydrophila reference strains.Pathogenic experiment showed that HBA-2 had a strong pathogenicity to mice and raccoon dog.Two virulence genes hlyA and epain HBA-2 were detected in four virulence genes aer,hlyA,epaand act by PCR method.The result of immune protection test showed that the isolated strain HBA-2 had good immunogenicity.The result of drug sensitivity test showed that HBA-2 was sensitived to enrofloxacin,ceftriaxone,amikacin,florfenicol and cefixime,and differently resistant to other drugs.
