摘  要:目的分析石家庄市2009-2016年甲型H1N1流感病毒分离株血凝素蛋白(HA)的基因特征,了解其变异情况。方法选取石家庄市2009-2016年甲型H1N1流感病毒分离株71株,PCR扩增HA基因并测序;利用生物信息学软件对分离株的HA基因进行进化和变异分析,并与北半球疫苗株A/California/07/2009进行对比;采用MEGA(5.05)软件NJ法构建进化树,进行系统进化分析。结果石家庄市71株分离株与疫苗株A/California/07/2009的HA基因核苷酸同源性为96.86%~99.57%,氨基酸同源性为96.77%~99.11%,株间核苷酸同源性为96.30%-100.00%,氨基酸同源性为96.95%-100.00%;进化树分为2支,部分2013年分离株及2014年和2016年分离株与疫苗株发生偏离;与A/California/07/2009相比,分离株HA蛋白有7处抗原位点和4个受体结合位点发生变异,NetNGlyc1.0预测8个糖基化位点均未发生缺失,2016年的4个分离株在162位新增一个糖基化位点。结论石家庄市甲型H1N1流感病毒分离株HA基因与疫苗株同源性逐年降低,氨基酸变异逐年增多,虽显示低致病性特征,但仍需继续对其加强监测。Objectives To analyze the genetic characteristics and variation of the hemagglutinin protein of influenza A(H1 N1)virus isolated in the City of Shijiazhuang from 2009 to 2016. Methods The hemagglutinin(HA)gene of 71 isolates of influenza A(H1 N1)collected in Shijiazhuang from 2009 to 2016 was amplified and sequenced.Bioinformatic software was used to analyze the evolution and variation of the HA gene.These strains were compared to the Northern Hemisphere vaccine strain A/California/07/2009,and an evolutionary tree was constructed via the NJ method using the software MEGA(5.05). Results Nucleic acids of the HA gene of 71 stains from Shijiazhuang were 96.86-99.57%similar to those of the vaccine strain A/California/07/2009.Amino acids of the HA gene were 96.77-99.11% similar between strains.Nucleic acid similarity between isolates was 96.30-100.00%,and amino acid similarity between isolates was 96.95-100.00%.The phylogenetic tree was divided into two clades.Some of the strains isolated in 2013,2014,and 2016 deviated from the vaccine strain.Compared to A/California/07/2009,the isolates had mutations at 7 antigenic sites and 4 receptor binding sites.NetNGlyc1.0 predicted that there were no deletions in any of the 8 glycosylation sites.Four isolates from 2016 exhibited glycosylation at 162 sites. Conclusion The HA gene of the influenza A(H1 N1)virus isolated from Shijiazhuang displayed less similarity to that of a vaccine strain each year.The variation in amino acids gradually increased each year.Although the virus has low pathogenicity,its HA gene needs to continue to be monitored.
