棣棠花的遗传多样性初探

摘  要:棣棠花是蔷薇科棣棠花属一种极具药用价值和观赏价值的野生植物资源,从8个叶绿体DNA序列和1个核DNA序列中筛选出1个有效DNA片段rps16,利用其对棣棠花6个居群58个个体进行了遗传多样性研究,以期为该物种的资源评价和利用奠定基础.结果显示:rps16序列矩阵长度为607bp,共检测到9个核苷酸变异位点和11个单倍型,居群总的遗传多样性(HT)为0.884,单倍型多态性(Hd)为0.778,核苷酸多态性(π)为0.001 2.棣棠花居群间遗传变异(62.84%)大于居群内遗传变异(37.16%),居群间变异是其主要变异来源;6个居群间具有较高的遗传分化(Gst=0.653,Nst=0.600,Fst=0.628 4),基因流水平较低(Nm=0.128 5).失配分析结果表明种群在进化过程中曾经历过快速扩张,中性检验支持上述结果:Tajima's D=-1.019 04(P>0.1);Fu and Li's D=-2.056 07(0.1>P>0.05);Fu and Li's F=-2.020 36(0.1>P>0.05).研究结果表明:棣棠花居群遗传多样性较高,居群间存在较高的遗传分化,奠基者效应和瓶颈效应可能是主要的影响因素.Kerria japonica (L.) DC., belonging to the genus Kerria DC. (Rosaceae), is an important wild plant resource with medicinal and ornamental value. In this study,the genetic diversity of Kerria japonica (L.) DC.were analyzed with chloroplast DNA and rDNA markers. Out of 9 candidate pairs, one effective molecular marker (rps 16) were screened,providing the molecular basis for further re search.The alligned sequences were 607 bp in length,and 9 mutation sites and 11 haplotypes were de tected.Tbe haplotype polymorphism ( Hd ) ,the nucleotide diversity(T) and the total genetic diversity (H T) were 0. 778, 0.001 2 and 0.884, respectively. The genetic variation among the population (62.84%) was higher than the intra population genetic variation (37.16% ), indicating that among population variation was the main source of variation of these populations. The genetic differentiation among the populations was high ( Gst =0.653, Nst =0.600, Fst = 0.628 4) and the gene flow was low ( Nm =0.128 5). Mismatch analysis revealed that the population had experienced rapid expansion dur ing the evolution process.and the neutrality test supported the above results:Tajima's D=-1.019 04( p 2〉 0.1);Fu and Li's D =-2.056 07(0.1〉 p 〉0.05); Fu and Li's F= -2.020 36(0.1〉 p 〉0.050). The resuhs showed that the genetic diversity of Kerria japonica (I..) DC. was high,and there is a high genetic differentiation among populations. The founder effect and bottleneck effect may be the main influencing factors.
