摘  要:目的:评估不同剂量蔗糖铁(iron sucrose,IS)通过耳缘静脉注射诱导家兔绝经后铁蓄积模型建立的可行性及对机制进行初步探讨。方法:普通级雌性家兔50只随机分为5组:假手术组、去势组以及模型A、B、C组,各10只。去势组和模型组行双侧卵巢切除术,假手术组仅切除卵巢周围部分脂肪组织后即关闭腹腔。术后模型A、B、C组分别通过耳缘静脉注射不同剂量IS(10、15、20 mg/kg),假手术组和去势组以同样方式和频次给予适量生理盐水注射。分别于造模前1 d和造模后第8周比较各组血清铁蛋白(serum ferritin,SF)和17β-雌二醇(estradiol,E2)水平,分析去势组SF与17β-E2的相关性。结果:造模后第8周,模型A、B、C组SF水平均高于假手术组和去势组,且模型C组均高于模型A、B组,比较差异均有统计学意义(P<0.01),去势组17β-E2水平低于假手术组(P<0.01),但两组SF水平比较差异均无统计学意义(P>0.05);去势组SF与17β-E2水平无相关性(r=-0.226,P=0.531)。结论:耳缘静脉注射IS可成功诱导家兔绝经后铁蓄积模型的建立,绝经后铁蓄积可能与机体铁排泄途径减少及摄入增多有关。Objective:To evaluate the feasibility of constructing the postmenopausal ion accumulation rabbit models induced through injection with various doses of iron sucrose(IS) via the ear vein,as well as to preliminarily probe into the mechanism.Method:50 normal grade female rabbits were randomly divided into five groups:sham operation group,castration group and model A,B and C groups,10 rabbits in each group. The castration group and model groups were performed bilateral oophorectomy,while sham group was conducted removal of part of the adipose tissue around the ovary,followed by abdomen closure immediately.The model A, B and C groups were injected with various doses of IS(10,15 and 20 mg/kg) via the ear vein postoperatively respectively,and sham operation group and castration group were administered injection with appropriate amount of saline with the same method and frequency.The levels of serum ferritin(SF) and 17β-estradiol(E2) in each group were compared at 1 d before modeling and eighth weeks after modeling,the correlation between SF and 17β-E2 in castration group was analyzed.Result:Eighth weeks after modeling,the levels of SF in model A,B and C groups were higher than those of sham operation group and castration group,and the model C group was higher than model A and B groups,the differences were statistically significant(P〈0.01),the level of 17β-E2 in castration group was lower than that of sham operation group(P〈0.01),but the SF level of two groups was compared,the difference was not statistically significant(P〉0.05).There was no correlation between SF and 17β-E2 level in castration group(r=-0.226,P=0.531).Conclusion:Injection with IS through the ear vein can induce the construction of the postmenopausal ion accumulation rabbit models,and the postmenopausal ion accumulation may be related to the reduced excretion pathways and increased intake of iron of the body.
