烟草提取液对大鼠Ⅱ型肺泡上皮细胞TGF-1β/smad2信号通路表达的影响及机制研究

摘  要:香烟烟雾(CS)是导致慢性阻塞性肺疾病(COPD)进展的主要危险因素,目前尚无有效的治疗方法来抑制该病的进展;同时,也有研究表明吸烟能够引起肺纤维化。本实验主要研究烟草提取液对大鼠Ⅱ型肺泡上皮细胞(ATⅡ)的影响及对其转化生长因子-β1/smad2(TGF-1β)信号通路的影响,并探讨其可能的作用机制。大鼠ATⅡ经4%烟草提取物处理后,TUNEL assay检测到大鼠ATⅡ数量减少并存在明显的凋亡小体,同时Western blot检测发现,TGF-1β蛋白表达量明显增多,p-Smad2蛋白表达量也出现了显著的变化。烟草提取物能够促进ATⅡ的凋亡,并且激活TGF-1β/smad2信号通路,本研究从TGF-1β/smad2信号通路的激活阐释了烟草提取物对ATⅡ损伤的可能机制,为以后的研究奠定了一定的基础。Cigarette smoke (CS) is a major risk factor for the development of chronic obstructive pulmonary disease (COPD), currently there is no effective treatment to prevent the progression of the disease; studies have shown that smoking can cause pulmonary fibrosis. This experiment mainly studied the effect of tobacco extract on rat type II alveolar epithelial cells (ATII) and its effect on TGF-1β / smad2 signaling pathway and explored its possible mechanism. After treated with 4% tobacco extract, ATII of rats were detected by TUNEL assay, the number of ATII was decreased and there was obvious apoptotic bodies, Western blot showed that the expression of TGF- significantly increased, p-Smad2 protein expression also showed a significant change. Tobacco extract can promote the apoptosis of ATII and activate the TGF-1β / smad2 signaling pathway, this study illustrates the possible effect of tobacco extract on ATII injury from the activation of TGF-1β / smad2 signaling pathway mechanism, which laid the foundation for future research.
