摘  要:目的:探讨多发性骨髓瘤骨髓组织中miR-181a表达量与肿瘤细胞增殖、迁移的相关性。方法:选择2012年3月~2017年12月间在本院确诊并治疗的多发性骨髓瘤患者52例作为多发性骨髓瘤组,另取同期在本院进行骨髓穿刺并最后明确骨髓功能无殊的健康者36例作为正常对照组。对比两组骨髓组织中miR-181a表达量以及疾病相关增殖基因、疾病相关侵袭基因表达量的差异,采用Pearson检验评估多发性骨髓瘤骨髓组织中miR-181a表达量与肿瘤细胞增殖、迁移的相关性。结果:多发性骨髓瘤组骨髓组织中miR-181a的表达量高于正常对照组;骨髓组织中PCDH10、TIP30 mRNA的表达量低于正常对照组,c-maf、ADAM10 mRNA的表达量高于正常对照组;骨髓组织中Annexin2、HSP90、PIK3R1、Twist1 mRNA的表达量高于正常对照组,TGFBI、PTEN mRNA的表达量低于正常对照组(P<0.05)。相关性分析发现,多发性骨髓瘤骨髓组织中miR-181a表达量与肿瘤细胞增殖及迁移活力直接相关。结论:多发性骨髓瘤骨髓组织中miR-181a表达量异常增高,具体表达量与肿瘤细胞的增殖及迁移活力直接相关。Objective:To investigate the correlation of miR-181a expression in bone marrow tissues of multiple myeloma with tumor cell proliferation and migration.Methods:52 patients with multiple myeloma who were diagnosed and treated in this hospital between March 2012 and December 2017 were selected as multiple myeloma group,and 36 healthy subjects who received bone marrow aspiration and were eventually proven to be with normal bone marrow function in this hospital during the same period were selected as normal control group.The differences in the expression of miR-181a,illness-associated proliferation genes and illness-associated invasion genes in bone marrow tissue were compared between the two groups,and Pearson test was adopted to evaluate the correlation of miR-181a expression in bone marrow tissues of multiple myeloma with tumor cell proliferation and migration.Results:miR-181a expression in bone marrow tissue of multiple myeloma group was higher than that of control group.PCDH10 and TIP30 mRNA expression in bone marrow tissues were lower than those of control group whereas c-maf and ADAM10 mRNA expression were higher than those of control group;Annexin2,HSP90,PIK3R1 and Twist1 mRNA expression in bone marrow tissues were higher than those of control group whereas TGFBI and PTEN mRNA expression were lower than those of control group(P〈0.05).Correlation analysis showed that the miR-181a expression in bone marrow tissues of multiple myeloma was directly correlated with tumor cell proliferation and migration activity.Conclusion:miR-181a expression significantly increases in bone marrow tissues of multiple myeloma,and the specific expression is directly correlated with the proliferation and migration activity of tumor cells.
