雷公藤内酯醇联合雷帕霉素抗人结肠癌细胞SW480增殖的体外研究

摘　　要：目的观察雷公藤内酯醇(Triptolide,TPL)和雷帕霉素(Rapamycin,RAPA)单用及联合应用对人结肠癌细胞SW480体外增殖的影响,初步探讨其作用机理。方法 MTT法检测TPL和RAPA单用和联合对人结肠癌细胞株SW480增殖的抑制作用;蛋白质印迹法测定IκBα、pIκBα、p-NF-κB及NF-κB蛋白表达。结果 TPL和RAPA联合应用对SW480细胞的增殖有协同抑制作用;蛋白质印迹法测定结果显示两药联合能够比单药显著抑制IκBα和NF-κB的磷酸化。结论实验证实TPL和RAPA能协同抑制SW480细胞增殖,其机制可能是通过抑制NF-κB信号通路而发挥作用。OBJECTIVE To observe the effect of TPL（Triptolide）and RAPA （Rapamycin）on the proliferation of human colon cancer cell line SW480 in vitro and its mechanism.METHODS MTT assay was used to detect the inhibitory effect of TPL and RAPA on the proliferation of human colon cancer cell line SW480 by single and combined use.The protein expressions of p-NF-κB and NF-κB were detected by Western blot. RESULTS The combination of TPL and RAPA could inhibit the proliferation of SW480 cells.The results of Western blot analysis showed that the combination of the two drugs could significantly inhibit the phosphorylation of IκBα and NF-κB.CONCLUSION The results showed that the combination of TPL and RAPA could synergistically inhibit the proliferation of SW480 cells,and its mechanism might be by inhibiting NF-κB signaling pathway.
